Neohesperidin dihydrochalcone(NHDC) is a highly efficient, non-toxic and low calorie sweetener, which can improve the flavor and inhibit the effect of bitterness at the same time, so it has broad application prospects in food industry. In this paper, we studied the preparation process of neohesperidin dihydrochalcone, and the product obtained through this process was characterized, then we studied how to improve the solubility and velocity in water of NHDC under the normal temperature.We chose naringin as raw material, according to certain proportions, after the process of hydrolysis in the alkaline conditions,the phloroacetophenone-4’-beta neohesperidin, an important intermediate product was obtained. At the basis of single factor experiment, we optimized the synthesis conditions by response surface design experiment. Then the phloroacetophenone-4’-beta neohesperidin was choosed as raw material, we found the optimum conditions by orthogonal design experiments. Finally we chose homemade new hesperidin as raw materials and synthesized the neohesperidin dihydrochalcone,which was anfinal product.Besides, The yield rate of NHDC was treated as a standard,we obtained optimal experimental conditions, and fusing point test,infrared analysis and H-NMR spectroscopy were used for analysis and identification for homemade products. Finally we discussed the modification application of NHDC, then in ordet to estimate the effect of modification on the function and application of NHDC, the solubility, dissolution rate,sweetness value and the effect of cover the bitter of these modificated products were measured, and the results are shown as follows:(1) Synthesis technique optimization of phloroacetophenone-4’-beta neohesperidinThe yield rate of phloroacetophenone-4’-beta neohesperidin was treated as a standard, we investigated the effects of sodium hydroxide concentration,response time, reaction temperature and liquid-to-solid ratio thses four factors on the yield rate of it.We optimized the results of single factor by response surface experiments,we found the best preparation techniques as follows:the amount of naringin was 15.0g, reaction liquid ratio was 11:1, amount of sodium hydroxide was 22.5 g, reaction time 2.08 h and the reaction temperature is 100℃. Under theses conditions, phloroacetophenone-4’- beta new hesperidin yield could up to 77.03%.(2) Synthesis and characterization of neohesperidin dihydrochalconeThe hesperidin yield was treated as standard, we investigated the effect of reaction time, amount of proline catalyst and amount of anhydrous ethanol on the yield of products,and we obtained the the optimal experimental through the orthogonal experimental design. The structure of the product was characterized by melting point determination, IR and NMR. The optimal crystallization conditions are as follows: reaction temperature was 80℃, Phloroacetophenone-4’- beta-neohesperidin content was 10g(0.021mol), the dosage of ethanol was 140 ml, reaction time was 10 h, amount of proline was 2.4g, amount of isovanillin was 3.6g, under this conditons,the highest yield of neohesperidin was 84.21%.The yield of NHDC was treated as a standard,we investigated the effects of reaction time, liquid material ratio, Na OH concentration and dosage of catalyst Pd/C on the yield of Neohesperidin dihydrochalcone, And we got the optimal experimental conditions through the orthogonal experimental design analysis.The structure of the product was characterized by melting point determination, IR and NMR. Experimental results are as follows: hesperidin content was 20 g, Na OH concentration was 1mol / L, amount of catalyst was 2.5g, liquid-solid ratio was 8:1, the optimal conditions reaction time was 12 h, under these conditions,the highest yield of NHDC could reached 81.83%.(3)Study of modification and application of neohesperidin dihydrochalconeThe NHDC was modified by mixing of Na HCO3, sodium cyclamate, sodium saccharin, amylose starch and beta-cyclodextrin. in ordet to estimate the effect of modification on the function and application of NHDC, the solubility, dissolution rate,sweetness value and the effect of cover the bitter of these modificated products were measured.The experimental results showed that compound modification could greatly improve the solubility and dissolution rate of the NHDC. The modificated NHDC ccould obtain many different sweetness specifications and masking bitter better.
