| Xiang A-2 Granules, which is made from Paeony、. Angelica and other seven kinds of herbs, is an effective prescription for preventing and curing AIDS. The author has made an experimental study on its preparation and quality standards.Objective Established the preparation and quality standards of Zipu Granules.Methods Using the content of paeoniflorin and extracting ratio as evaluating indexes, we selected the optimum extracting condition with water by orthogonal design. On the basis of the one-factor investigation such as the dosage of chitosan, the concentration and pH of the extracting solution, temperature and stirring etc, the orthogonal design was selected to measure the effects of the chitosan dosage,the concentration and PH of the extract solution on the flocculation process.A lock of tests has been made to select the optimum condition of concentration and frying factor,preparative technology and suitable auxiliarant. Angelica、Hedyotis diffusa were indentified by TLC. For the sake of assay of paeoniflorin, a new high performaliquid chromatography method was developed. Results The optimum extracting condition was:soaking in water for 0.5 h, extracting two times,adding water for 12, 10times and extracting for 2.5, 2.0 h, respectively. The optimal flocculation process conditions were that the adding dosage of chitosan was 0.6mL·g-1, the concentration of meterials vs extract solution was 1:4,the PH was 7,the flocculation temperature was 60℃,the stirring speed was 200r·min-1 and the stirring time was 10 minutes. The spray dry temperature is 60℃. On the basis of these studies combining specifity of medicinal materials and requirements of the dosage form, the preparation process was ascertained for the Xiang A-2 Granules. Angelica% Hedyotis diffusa、Buplearum and Zhejiang pui were indentified by TLC. The average recovery rate was 100.76% while RSD was 2.39%.Conclusion The procedure was reasonable,stable and practical,the quality standards was set up. |
