Huoxueantong Granule, which is made from Radix Paeoniae Rubra、Radix Dipsaci、Radix Angelicae sinensis and Radix aconiti Kusnezoffii and so on, is an effective prescription in treating Rheumatoid arthritis、Joint pain of Lumbar and Leg. It has analgesic effect on old wounds too.The therapeutic effect is to have Huoxue analgesia and reduce inflammation and remove Rheumatoid. We have made a systematic study its preparation and quality standards, Therefore, it provides the scientific proof for its clinic effect.1.Study on the preparationStudy on the extract process of volatile oil、the process of the beta-cyclodextrin Inclusion technology for volatile oil、the extract process with ethanol、the concentration process、the drying process and the molding process. Finally it was decided①The optimal conditions of the extract process of volatile oil as follows:1 hour of soaking,8 times of water-adding amount, Heating reflux extraction with water by 8 hours.②The best conditions of the process of the beta-cyclodextrin Inclusion technology for volatile oil as follows:the proportion of③β-CD and volatile oil was 8:1,4 times of water-adding amount, inclusion time was 3h.③Alcohol Extraction Process:0.5 hours of soaking,2 times of refluxing with 60% alcohol,12、10 times of alcohol-adding amount and 2.0,1.5 hours of refluxing time respectively.④Study of the concentration and desiccation:decompression concentration, The physic liquor was vacuum dried at 60℃, the vacuity was-0.08Mpa, the extracta sicca was easy to crush to pieces.⑤Study of the molding process:the molding process choose lactose and 90% alcohol.2.Study of quality standardsRadix Paeoniae Rubra、Radix Dipsaci、Radix Angelicae sinensis、Radix et Rhizoma salviae Miltiorrhizae、Ramulus Cinnamomi、Rhizoma Atractylodis、Herba Visci、Flos Carthami and Aconitine of Radix aconiti Kusnezoffii were identified respectively by TLC. The content of Paeoniflorin and AsperosaponinⅥwere determined in Huoxueantong Granule by HPLC.In the results, average recovery rate of Paeoniflorin was 98.43%, while RSD was 1.93% and average recovery rate of AsperosaponinⅥwas 97.2%, while RSD was 1.36%. the method was reliable、precise and can be applied for quality control.
