3D microscope plays a key role in many fields. Currently, due to the conventional algorithm, a high precision linear stage with piezo actuator is implemented commonly and dominates the cost of the microscope. The conventional algorithm for determining the contrast value of the focal depth response requires accurate phase shift of the fringe pattern illumination and many image acquisitions. Now a new algorithm, which is robust to inaccurate phase shifting of the fringe pattern illumination, is developed by Heilbronn University, which can remain the accuracy with inaccurate phase shift and reduce the image acquisitions.This master dissertation is based on the new algorithm and the aim is to design a low-cost micro-motion system driven by a common motor for testing the new algorithm. Designing a cam-follower mechanism actuated by a stepper motor will realize the sys-tem and satisfies the requirements without reducing the accuracy. The concrete research contents and results are as follows.1. Based on the available space of the3D microscope, the mechanical structure design has been realized through a cam-follower system, which is guided by an elastic guide mechanism and can meet all the requirements for image acquisitions.2. A Graphic User Interface has been generated to control the stepper motor corpo-rate with the camera and the stage motor.3. The experiment of controlling the new designed mechanical system has been done and the experimental sampling distance has been calibrated and optimized. Further, the experiment of testing the new algorithm with the new designed micro-motion mechanism has been done as well. The experimental results show that this micro-motion mechanism can satisfy the requirement of replacing the piezo-actuated high precision linear stage without reducing the accuracy. Moreover, reducing the cost of3D microscope is realized as well.
