With the development of the nuclear energy and industry,as well as the threat of nuclear terrorist attacks,the risk of radiation exposure to public is increasing. A fast,high-throughput and convenient biodosimetry is of great importance in response to the unplanned exposures to ionizing radiation,such as nuclear reactor accident. Early biodosimetry has therefore become a research hotspot since people realized the importance of biodosimetry. Change of protein based biodosimetry attracted of people’s attentions.Connexin43(CX43)and Clusterin(CLU) protein in mice serum were detected to investigate their variation with time and dose after a acute whole-body 60 Co γ irradiation. And to validate their practicability to be a new biodosimetry. Two conclusions gained below:(1) The content of CX43 protein in mice serum was detected using surface plasmon resonance(SPR) technology.SPR biological sensing chip coated with CX43 antibody can specifically detect the CX43 standards and the CX43 proteins in samples.The detection limit of CX43 standard is as low as 5ng/L, and SPR technique is more sensitive compared to ELISA technique. Obvious up-regulation of CX43 expression was observed by γ-rays doses as lows as 0.6 Gy. CX43 protein in mice serum first increased and then decreased with the time after mice were irradiated by a dose of 1.2 Gy. The highest level of CX43 appeared 3 hours after irradiation. At this moment, the content of CX43 showed a linear correlation with the γ-rays doses in the range of 0~2.4 Gy, however it began to reduce when the dose continued to increase. Western blot results showed the same results with SPR sensor. Detection of Connexin43 protein in mice serum using SPR technology is expected to be a new biodosimetry.(2) The content of CLU protein in mice serum was detected using one-step ELISA method, and its variation with time and dose after a acute whole-body 60 Co γ irradiation was studied. Time effect results showed that, CLU protein in mice serum first increased and then decreased with the time after mice were irradiated by a dose of 1 Gy. It reached to top at 2 h and then it decreased with time. At 2 h, the content of CLU showed a linear correlation with the γ-rays doses in the range of 0~1 Gy, however it began to reduce when the dose continued to increase. Detection of CLU protein in mice serum using one-step ELISA method has the potential to be an early,low-dose biodosimetry.
