Based on fingerprint technique and taking reference from chemical quality control model, the establishment of new Chinese Traditional Medicine (CTM) quality standard, or CTM fingerprint has been recognized as the most effective way for TCM quality control management at present. Fingerprint based on ultra high performance liquid chromatography (UPLC) is a new technique for TCM analysis and quality control, apart from the strength of time-efficiency for research and less reagent needed for the test; it also has higher definition and accuracy than high performance liquid chromatography (HPLC), which is better for CTM analysis and quality control. Rauwolfia is an important hypotensor, an establishment of fingerprint able to reflect the intrinsic chemical properties at this stage, can better indicate the intrinsic quality of Rauwolfia, upgrade the technological level for Rauwolfia quality evaluation and provide scientific basis for Rauwolfia quality control. Meanwhile globally UPLC is a new technique for analysis, which is’ still at the early stage for medicinal application, there’s very few domestic report about CTM quantitative and qualitative analysis and building fingerprint using UPLC-PDA and UPLC-MS. It’s hopeful to enrich the experience of UPLC application in CTM research through study on Rauwolfia UPLC fingerprint.This Paper consists of 4 chaptersChapter 1, Introduction of the research progress of CTM fingerprint, application of ultra high performance liquid chromatography (UPLC) in CTM fingerprint and CTM quality control, and outline for RauwolfiaChapter 2, separation analysis for methanolic extract from Yunnan Rauwolfia by UPLC associating with mass spectrometry (MS), realizing the 8 important separations for reserpine、yohimbine、Serpentinine、ajmalicine、ajmaline、rescinnamine、raunescine and Deserpidine mentioned in the references, which benefits the peak belongingness research in Rauwolfia UPLC fingerprintChapter 3, according to related parameters from fingerprint of Yunnan Rauwolfia from different regions, identified 42 peaks; assort the peaks by number of 1 to 42 from low to high according to the average value of retention time, among them there’re 22 common peaks found totally. By comparing common peaks respectively in different ways, similarities were all found between 0.7 to 1.0, part of them below 0.9 though, which indicated that certain differences existed between Yunnan Rauwolfia from different regions. Through main constituent analysis on common peaks’area percentage, the differences of samples were reflected by peak 1、3、5、6、12、15、22、28、29,32 and change of dissolving rate. This result showed a perfect quality control has to be achieved by base training building for Yunnan Rauwolfia, picking improved breeds according to actual needs, establishing supply channels for genuine medicinal herbs.According to related parameters in fingerprint of Rauwolfia from different regions,42 peaks were identified; assort the peaks by number of 1 to 42 from low to high according to the average value of retention time, among them there’re 22 common peaks found totally. By comparing common peaks respectively in different ways, similarities for both emetic and Hainan Rauwolfia were lower than 0.1, similarities of other Rauwolfias ranged between 0.8 to 1.0. This result showed significant differences separated emetic and Hainan Rauwolfia from other Rauwolfias. Cluster Analysis for common peak area normalization also verified this conclusion.Association with mass spectrometry (MS), identified parts of the chemical composition belongingness in UPLC fingerprint, estimating peak 39、42、35、29、34、41、28、40、38 was rauvanine、Aricine、purpeline、vomilenine、reserpine、ajmalicine、yohimbine、rescinnamine、Serpentinin respectively.Chapter 4, on the basis of Rauwolfia fingerprint research, established ways of determinations for amounts of yohimbine, reserpine,ajmalicine in Rauwolfia simultaneously by UPLC, used UPLC to measure amounts for yohimbine, reserpine and ajmalicine from different kind of Rauwolfia as well as comparative study on the amounts. The result was:the amount of yohimbine, reserpine and ajmalicine in Rauwolfia roots was thick root>medium root>thin root; the biggest amount of reserpine in the Rauwolfia root extract was from Hainan Rauwolfia, the biggest amount of yohimbine in the Rauwolfia root extract was from Yunnan Rauwolfia from botanical garden of Chinese Academy of Sciences in Xishuangbanna, the biggest amount of ajmalicine in the Rauwolfia root extract was from Guangdong Rauwolfia, the amount of reserpine in root extract of emetic Rauwolfia was much higher than the one found in Yunnan Rauwolfia, the amount of yohimbine in root extract of emetic Rauwolfia was much lower than the one found in Yunnan Rauwolfia...
