Characterization Of An Improved Strain (SF-2) Of Porphyra Haitanensis (Bangiales, Rhodophyta) And Its Pilot Cultivation In Mariculture Farm

The characteristics of the improved strain SF-2 were evaluated and compared with the improved variety SF-1 and the wild type of Porphyra haitanensis according to the laboratory studies and pilot scale tests in mariculture farm., such as growth, content of photosynthetic pigments and phycobiliprotein, thickness of the blades, high temperature resistance, numbers of released conchospores and yield in pilot cultivation.In lab study, we found that the SF-2 strain had a long growth period with a absolutely growth rate about 1.5 times larger than the wild type .After 100 days cultivation, the SF-2 strain still had a fast growth rate as well as a length of 390 cm which was 10 times longer than the wild type. However, the SF-1 strain had a similar growth tendency to the SF-2 strain. In the wavelength of 350-750 nm, in vivo absorption spectra of the blade of SF-2 strain showed five absorption peaks as well as the wild type and the SF-2 strain, however, the blade of SF-2 strain showed higher absorption value in every peak than the wild type. The blade of SF-2 strain had a photosynthetic pigments content of 8.98mg/g and a phycobiliprotein content (PE+PC) of 90.8 mg/g, which were 33.2% and 95% increased than the wild type respectively, but a little more than the content of SF-1 strain . Mean thickness of the blade of SF-2 strain was 31.95um, which decreased 38.7% and 14.0% compared to the wild type and SF-1 strain, respectively. We also found that the SF-2 strain had an excellent high temperature resistance than the other two strains at 29±1℃.While cultured in mariculture farm, the SF-2 strain had a number of released conchospers of 286 thousand per shell, which increased 43.0% than SF-1. We collect four harvests of these three strains(breed) inmariculture farm, and found that SF-2 had the highest yield, and the mean yield per unit area was up to 1600 kg, which increased 30.1% and 6.7% than WT and SF-1 strains, respectively. In the wavelength of 350 ~750 nm, in vivo absorption spectra of the blade of SF-2 also showed five absorption peaks as the laboratory study, the blades of SF-2 showed higher absorption value in every peak than WT and SF-1 strains, and the P5 absorption peaks of the blades of SF-1 were similar to WT, while the others higner than WT. The content of photosynthetic pigments of the blades of SF-2 cultured in mariculture farm was 20% increased than that of WT and SF-1 strains, while the phycobiliprotein content (PE+PC) was 55.8% and 22.3% increased than WT and SF-1, respectively. And the thickness of the blades showed a similar result to the laboratory study.It came to a conclusion that the superiority of the improved strain SF-2 possesses the value to be applied and disseminated.
