The genus Lilium consists of about 55 species distributed in China, most of them only living in China. In recent years, lots of researches of genetic diversity about this genus were fastened on morphological traits,cytology,isozyme and DNA molecular makers,however, it is not well known in the middle reach of Yangtze river.In this study, we used the technique of ISSR molecular makers to analyse the genetic relationship among wild germplasms here, meanwhile, investigated population genetic structure of L.leucanthum and L.brownii, in order to provide a basis for wild germplasm identification, breeding and formulating conservation strategies for this ragion.The main results were obtained as follows:Preliminary observation about growth from three kinds of Lilium. There were obvious different between phenophase and growth in different species The difference of growth was little in different populations. L. lancifolium bud period earliest and the flowering period was as long as forty-three days in three kinds of Lilium It seemed that L. lancifolium had better ornamental value and utilization potential. The correlation was the strongest between flowering and bulb quality. Research genetic relationship of Lilium wild germplasm(1) 11 ISSR primers were used to investigate the genetic relationship among 269 wild Lilium germplasm from 8 different regions of China.In total,117 bands were amplified by 11 ISSR primers,resulting 100% polymorphism at genus level, and the avenue value of SM similarity coefficient was 0.58.(2) The UPGMA dendogram showed there were closely relationship between L. leucanthum and L. brownie; L. concolor was much closer to Section Leucolirion; The close relatedness among L. speciosum var. gloriosoides、L. rosthornii、L. henryi and L.×formolongo.(3) Clustering analysis data revealed that the clustering pattern of Lilium germplasm was tightly linked with their geographical origins. The evaluation of genetic structure of 4 populations of L. leucanthum(1)L. leucanthum possessed fairly high degree of genetic diversity at species level,the values of PPL,h and I were 98.31%,0.3786 and 0.5532 respectively. The genetic diversity sequence was CWP>CWJ>HB>SNJ. (2) The genetic identity varied from 0.9063 to 0.9541 among 4 populations; and the genetic distance varied from 0.0470 to 0.0984.4 populations were separated into 2 groups with relation to geographical origins, namely, CWJ-. CWP and HB clustered into one group. SNJ population was clearly separated from other populations. Genetic differentiation among population of L. leucanthum was related to their longitude.(3) The AMOVA analysis indicated 93.89% of the total variance resided within the populations, and that only 6.32% of the variation resided among populations. The results displayed that conspicuous genetic differentiation existed within populations. The evaluation of genetic structure of 6 populations of L.brownii(1) At L. brownii species level,the values of PPL,h and I were 98.87%、0.3800 and 0.5555 respectively which were higher than L. leucanthum. Of the 6 populations, the sequence of genetic diversity was CWL>SNJ>ZG>BD>CWJ>YC.(2) The genetic identity varied from 0.9083 to 0.9677 among 6 populations; and shared the small genetic distance was 0.0328-0.0961. The genetic distance between HB and ZG was the nearest in the six populations.The following was Wushan populations and Yichang populations There was the farthest distance with SNJ populations. Genetic differentiation among population of L.brownii was related to their north latitude and geographical distance..(3) The AMOVA analysis showed 92.03% of the total variance resided within L.brownii populations, and that only 7.97% of the variation resided among populations. That is to say, a majority of genetic differentiation was found within populations.(4) Coefficient of genetic differentiation(Gst) was 0.1388 and 0.1263 respectively among L.leucanthum and L.brownii, the ene flow was 3.1013 and 3.4587.Geographical distance on the influence of yichang lily gene flow is remarkable, presumably with plant gene flow distribution scope of.Geographical distance had a significant influence for Nm of L. leucanthum. Maybe there was a close relationship between Nm and Plant distribution.
