Japanese Encephalitis Virus(JEV) was an important pathogen in reproductive disturbance of sows, and often mixed infection with many viruses, it could bring a lot of disadvantage to development of pig industry, it is very important to strengthen the research on JEV.1By quantitative real-time PCR the JEV RNA levels and gene expression levels of5cellular genes(Bcl-2、Bcl-xl、FasL、p53and PBR)of lymphocytes were detected after infection with Japanese Encephalitis Virus, result after an hour with JEV infected, the virus DNA can be detected, after72hours with JEV infected, the peak value was16times, Bcl-2began to express after1hour, with JEV infected and it could reach large number expression in3hours, then decrease, in72hours it was increased to a peak value; after1hour with JEV infected、3hours、24hours、72hours, FasL was increased1.25times、2.25times、4.8times and12.2times, its expression was decreased in12hours、48hours; the expression of Bcl-xl was decreased after JEV infection1hour、12hours、24hours、48hours, its expression reached4.8ti mes、560times in3hours、72hours; expression of p53was not change after JEV infection1hour、3hours、12hours、24hours、48hours, in72hours its expression amount was reached a maximum value and the expression amount could reach15times; expression of PBR could reach1.5times,7.7times,9times,82times after JEV infection1hour、3hours、12hours、72hours and its expression was decreased in24hours、48hours.2By the use of real-time quantitative PCR in lymphocytes with JEV infected in IL-8、12p35、12p40、IL-17、IL-18,the results showed that the expression amount of IL-8could reach1.7times、10times、3.2times、2.8times after JEV infection1hour、3hours、24hours、72hours, its expression was decreased in12hours、48hours; the expression of12p40could reach1.6times、5.7times、5.3times and1.9times after JEV infection1hour、3hours、12hours、24hours, and then its expression decreased in48 hours, even in72hours it could not express;12p35began to express in infection1hour, and the expression was decreased; its expression were increased after3hours、12hours、24hours, its expression could reach a peak in48hours, about its expression amount could reach73times, the expression was decreased in72hours; the expression of IL-17was not change after JEV infection1hour、3hours、12hours and expression was not change after24hours、72hours, in48hours its expression could reach a large number of expression; IL-18began to express in1hour, it could reach large number expression after JEV infection. its expression was increased after3hours、12hours、24hours、48hours; its expression could reach a peak in72hours and about7.8times.3By the use of real-time quantitative PCR with JEV infected in lymphocyte cells in MIP-la、TGF-β1、PGK1, the results showed that the expression of MIP-1a began to express with JEV infected in1hour, it could reach a lot of expression after3hours、12hours、24hours; it was no expression in48hours, its expression could reach a peak in72hours; The beta of TGF-β1was increased1.1times、1.09times、3.6times after JEV infection1hour、3hours、72hours, its expression was reduced after12hours、24hours、48hours; PGK1was not change after JEV infection1hour、12hours、24hours, the expression was decreased in3hours、48hours, it could reach large number expression in72hours.In summary, we could do the research of JEV peripheral blood lymphocyte cytokines transcription phase of swine, in order to grasp cytokines immunological function and JEV molecular mechanism, and theoretical basis is important for the prevention and treatment of JEV.
