| The immune system is the important system for the body to perform immune response and immune function.One study showed that the heritability of the body’s immune response was 25%.This heritability is similar with heritability of production performance and the shape.The higher immune response ability of bulls could be made full use,which improved the utilization rate of breeding bull cattle,enhanced generally herd immunity.And the higher genetic immune responses can be passed on to offspring.The excellent Holstein young sires were used to study cellular immunity and antibody immune in this paper.The flow cytometry,fluorescence qPCR,lymphocyte proliferation and ELISA liquid blocking antibody testing experiment method were taken to analyze the ability of the immune response among different individuals.The young sires were divided into low and high immune response individuals based on immune response level,and the differences were analyzed between individuals with high immune response ability and low immune response ability.The humoral immunity test was performed to detect serum antibody levels in different individuals after immune stimulation by ELISA liquid blocking antibody testing.The results showed that there was significant difference among different individuals at different time.Antibody titer of the higher young sires appeared significantly decreased after maternal antibody stimulation.Screening of high antibody mediated immune response by population antibody titer.The antibody titer of type A was obviously different by comparing three higher subtypes antibody titer and the lower ones in the foot and mouth disease during different stages of the immune stimulation.There is no significant difference on expression rate of before and after herd immunity through the T lymphocyte subgroup CD4~+CD8~+ detection.But there was significant difference among different individuals after ConA stimulation.The results showed that the individuals with higher proliferation rate had higher ability of immune response,and the lower proliferation rate.Fluorescent quantitative PCR assay was used to determine the expression of Th1 type cytokines IL-2,IFN-and Th2 cytokines IL-4 and IL-6.T cells mitogen ConA and B cells were stimulated by mitogen LPS respectively.The results showed that there were significant differences in the expression of immune factors among different individuals.There was nosignificant difference between ConA and LPS.The higher and lower individuals were screened immune response by comprehensive analysis of the cellular immune response ability among individuals.In this study,Mycoplasma bovis was used as the infection pathogen,and the group of the high and low free group were verified.The conclusion drew from that,mycoplasma could interfere with the host immune response ability,moreover,the disturbance of mycoplasma infection to host immune response was weak for the group with high immunity,and powerful for the group with low immunity. |
