Influence Of Reticuloendotheliosis Virus Infection On T Lymphocyte Subtypes And Their Related Cytokines In Peripheral Blood Of SPF Chicks

Reticuloendotheliosis virus(REV)can cause severe immunosuppression in infected poultry and bring great economic losses to poultry industry all over the world.The virus reaches the target organs mainly through the blood in the body.Although erythrocyte is the main cell type in peripheral blood,other immune cells also play an important role in anti-viral infection.In this study,1-day-old SPF chicks ’ blood were taken from the hearts to separate serum and lymphocyte.At 1,3,7,14 and 21 days after infection,the T lymphocyte subtypes(including CD4+,CD8+ T lymphocyte number and its ratio)in peripheral blood,IFN-γ,TNF-α,IL-2,IL-4,IL-10 and IL-12 content were detected by FACS and ELISA.In addition,the Th1/Th2 ratio was calculated according to the content of IFN-γ and IL-4.The main results are as follows:(1)The number of CD4+ T lymphocytes in peripheral blood of one-day-old SPF chicks infected with REV was significantly(P < 0.05)or extremely significant(P < 0.01)lower than that of control chicks at 1 to 21 days.The number of CD8+ T lymphocytes in peripheral blood of one-day-old SPF chicks infected with REV was significantly(P < 0.05)or extremely significant(P < 0.01)lower than that of control chicks at 1,3,14,21 days,but it was extremely significant higher(P < 0.01)than that of control chicks at 7 days.The ratio of CD4+ / CD8+ T lymphocytes was significantly higher(P < 0.05)than that of control chicks at 3 days and significantly lower(P < 0.05)or extremely significant(P < 0.01)than that of the control chicks at 7 to 14 days after virus infection,no significant difference(P>0.05)in the other times.The results showed that the number of CD4+ and CD8+ T lymphocyte subtypes in peripheral blood of chicks and the ratio between them were decreased because of the infection of REV,and then the immune function of chicks infected with REV were led to a low state.(2)In peripheral blood of one-day-old SPF chicks infected with REV,the content of IFN-γ were extremely significant(P < 0.01)lower than that of control chicks at 3 and 14 days,and extremely significant(P < 0.01)higher than that of control chicks at 7 days,the content of TNF-α were significant(P < 0.05)or extremely significant(P < 0.01)lower than that of control chicks at 3 and 14 days,and significant(P < 0.05)higher than that of control chicks at 7 days,the content of IL-2 were significant(P < 0.05)or extremely significant(P < 0.01)lower than that of control chicks at 1 to 14 days,the content of IL-12 were extremely significant(P < 0.01)lower than that of control chicks at 3 days,no significant difference(P>0.05)in the other times.The results showed that the biosynthesis and release of inflammatory factors could be promoted because of REV infection.(3)In peripheral blood of one-day-old SPF chicks infected with REV,the content of IL-4 were extremely significant(P < 0.01)higher than that of control chicks at 3 to 7 days,the content of IL-10 were significant(P < 0.05)or extremely significant(P < 0.01)higher than that of control chicks at 3 to 14 days,no significant difference(P>0.05)in the other times.The results showed that the infection of REV not only inhibited the biosynthesis and release of inflammatory factors,but also promoted the anti-inflammatory factors.(4)In peripheral blood of one-day-old SPF chicks infected with REV,the Th1/Th2 ratio were significant(P < 0.05)or extremely significant(P < 0.01)lower than that of control chicks at 3 and 14 days,but significant(P < 0.05)higher than that of control chicks,no significant difference(P>0.05)in the other times.The changes of major T lymphocyte subtypes and related cytokines in peripheral blood of SPF chicks infected with REV were systematically described in thes study.The effect of REV on the immune fuction of T lymphocytes in peripheral blood of SPF chicks was revealed,which provide a new thought for the prevention and treatment of RE.
