Porcine rotavirus is double-stranded RNA viruses, which mainly affect the function of theintestinal mucosa, resulting in malabsorption of nutrients, thereby causing diarrhea. The innateimmunity and adaptive immunity are connected by intestinal mucosal immunity, but mucosalimmune mechanisms that resist infection by PRV is not yet clear. Lots of studies have shownthat dendritic cells play an important role in the immune response. It is an important antigenpresenting cells, and it plays a key role to regulate the innate and adaptive immunity. Whentaking further research to DCs, we will understand the mechanism of the immune responseagainst viral infections. It is conducive to prevent and treat diarrhea that caused by PRV. Whichwill provide theoretical guidance to prevention and treatment for PRV disease.In this study, there are the number of162,4-5week-old BALB/c mices, were randomlydivided into three groups, PRV group, Poly (I:C) group and the PBS control group. Theexpression of CD11c+CD11b+CD8α+DC, CD11c+CD11b+CD8α-DC, CD11c+B220+CD8α+DC,CD11c+B220+CD8α-DC and MHC-Ⅱ, CD80, CD86, CD40of spleen, MLN, PPs of theBALB/c mices that infect PRV and Poly (I: C) by FCM in different times. Results thatcompared wih PBS showed that DCs of spleen, MLN and PPs of infecting by PRV become tobe activated and differentiated to CD11c+CD11b+CD8α+DC, CD11c+CD11b+CD8α-DC,CD11c+B220+CD8α+DC, CD11c+B220+CD8α-DC. The expression levels of MHC-Ⅱ, CD80,CD86and CD40of DCs are increase, which show that DCs have matured. The differentiation ofCD11c+CD11b+CD8α+DC and CD11c+CD11b+CD8α-DC of MLN are more obvious than spleenand PPs, CD11c+CD11b+CD8α-DC began to differentiate in12h, the differentiation levels ofCD11c+CD11b+CD8α-DC is more than CD11c+CD11b+CD8α+DC, and the differentiation levelsof CD11c+CD11b+CD8α+DC reached the highest level in120h. The differentiation levels ofCD11c+B220+CD8α+DC and CD11c+B220+CD8α-DC of spleen and MLN is the highest in72h,PPs in48h. The expression levels of MHC-Ⅱ, CD80, CD86, CD40reached the highest level in48h,72h,168h of the three organizations. The differentiation of CD11c+CD11b+CD8α+DC,CD11c+CD11b+CD8α-DC, CD11c+B220+CD8α+DC of the three organizations of infecting byPoly (I:C) did not significantly to PRV, but the differentiation of CD11c+B220+CD8α-DC ishigher than the PRV; wherein the expression of MHC-Ⅱ, CD80, CD40is increased, but littlechange in the expression of CD86. By summing up the results of the study showed that micesinfected by PRV inducing DCs of spleen, MLN and PPs are activated and are turn to mature. Thus induce an immune response. Poly (I:C) are different with PRV. It indicate that PRVinfection take the DCs turn to mature to inducte immune response, in the process not only withthe TLR3signaling pathway, as well as other factors.
