The vaginal bacterial flora plays a crucial role in the maintenance of healthy status of cow’sgenital tract.The non-toxic and no residue probiotics have been an important method to modifythe micro-ecological environment in cow’s genital tract and cure the cow’s genital tractdisease.In this study15dairy’s mucus samples of30-60days postpartum healthy Holstein cowswhich near the mout of the uterus were collected by endometrial biopsy sampler made by ourlaboratory. After cultured, separated and puruficated by traditional training methods,47Gram-positive bacteria were obtained.After the antibacterial effect of the isolated strains were tested, four strains were find havean antibacterial effect against the pathogenic E.coli, numbered1#,3#,5#and9#, and itsinhibition zone was11.60mm,10.77mm,3.57mm and1.13mm respectively. The antibacterialeffect of1#,3#against E.coli was significantly higher than that of5#(p<0.05), and Theantibacterial effect of1#,3#against E.coli was also significantly higher than that of9#(p<0.01).The four strains were identified by acteriological identification and16S rDNA molecularbiology techniques,the results shows that1#is Weissella koreensis,3#is Enterococcus hirae,5#Leuconostoc Lactis,9#is Ilyobacter polytropus.Then the growth performance, acid production performance, H2O2production property andthe mixed culture’s bacteriostatic action were tested. The results show that1#and9#strains arethe growing fastest strains and the production of the two acid can also to be the best, the9#’sH2O2production property is the strongest among others, the mixture of3#,5#and9#’sbacteriostatic action against E.coli is higher than the other mixed group(p<0.05), thebacteriostatic ring diameter are13.65mm and13.33mm respectively.The hydrophobic surface properties of the4strains were tested by cetane, toluene andxylene.The results show the hydrophobic effect of1#,3#and5#against these three kinds ofhydrophobic solvent was not significant(p>0.05), compared with9#, the hydrophobic effect of1#,3#and5#against these three kinds of hydrophobic solvent were significant differences(p<0.05). Cell adhesion experiments show that the average number of9#bacteria adhered onthe cow vaginal epithelial cells reached to22.52bacteria/cell,and1#,3#,5#were13.72bacteria/cell,17.52bacteria/cell and19.76bacteria/cell,respectively. The mixed strains’ of1#,3#adhesion results show that the average number of bacteria were35.60bacteria/cell,1#mixedwith9#were27.00bacteria/cell. The four strains of bacteria adhesion are positively related with hydrophobic effect.In conclusion,we combined the traditional bacterial culture appraisal method and molecularidentification methods,and4lactobacillus were isolated and identificated successfully,which canbe used as probiotics to prevent and cure cow’s genital tract diseases.
