Microorganism is closely correlated with the organisms' health and physiology.In recent years,the effect of genital tract microorganism in human health and disease has been paid more and more attention.Some researches showed that genital tract microorganism was affected by hormones.And the pregnancy situation and the reproductive performance are related to the host's secretion of hormones.Therefore,we supposed that there exist some differences in reproductive tract microorganisms related to pregnancy situation and reproductive performance.The pig is a multiparous mammal.And litter size is one of the most important economic traits for them.To improve litter size of pigs is of great significance.Microbes have been hypothesized to be involved in the physiology and pathophysiology of assisted reproduction since the first success in IVF,but it is still unknown for the possibility of reproductive tract microbiome associated with litter size.And Erhualian sow is known for litter size in the Taihu Lake area,but we found obvious difference of litter size in Erhualian sows.This experiment attempts to research the differenciation of microflora in genital tract of high or low prolificacy sows in pregnancy or non-pregnancy with Erhualian pigs model by high-throughput sequencing,whether microorganisms associated with the performance.Experiment 1 Measurement of FSH,E2 and P4 of high and low prolificacy Erhualian sowsErhualian pigs have high reproductive performance,but variation of litter size was found.In this experiment,based on EBV of TNB(EBVTNB)from 177 sows,9 high prolificacy and 9 low prolificacy Erhualian sows with similar age and parity were selected.After estrus synchronization and artificial insemination,18 sows were slaughtered to collect blood and uterine flushing fluid for follicle stimulating hormone(FSH),estradiol(E2),progesterone(P4),and other reproductive hormones detection on the 13 th day of breeding at the second natural estrus cycle.The pregnancy and prolificacy didn't effect the FSH,E2 and P4 in serum,but the E2 of serum was effect by the interactions of pregnancy and prolificacy(P<0.05).The P4 of serum tended to increase in pregnancy sows(P = 0.097).The E2 in uterus rinses was influenced by the state of pregnancy(P<0.05).Others had no significant differences.Experiment 2 Differenciation of microflora in vagina and cervix of high and low prolificacy sowsAt the phylum level,Proteobacteria and Bacteroidetes were dominant phyla in different litter size and different pregnancy situation in Erhualian sows.Acidobacteria in vagina were significantly influenced by pregnancy(P<0.01).Acidobacteria was tended to increase in vagina of low prolificacy sows(P = 0.079).Actinobacteria was tended to increase in vagina of high prolificacy sows(P = 0.075).The pregnancy physiological state and prolificacy of sows significantly impacted on Firmicutes in vagina(P<0.05).The pregnancy physiological state and prolificacy did not effect the microorganisms in cervix.But the interactions of pregnancy and prolificacy significantly effected Bacteroidetes(P<0.05).At the genus level,Pseudidiomarina had a tendency to rise in vagina of high prolificacy sows(P = 0.079).The relative abundance of Marinimicrobium in vagina was significantly effected in pregnancy group(P<0.05).Georgenia in vagina was effect by the interactions of pregnancy and prolificacy(P<0.05).Acinetobacter in cervix of high prolificacy sows were significant higher than low prolificacy sows(P<0.05).And Luteimonas in cervix of pregnancy sows were significantly higher than non-pregnancy sows(P<0.05).In conclusion,high and low prolificacy Erhualian sows in pregnancy or non-pregnancy pigs significantly effected the structure and composition of vagina and cervix microbial flora.
