This paper determined the growth of Vaccaria segetalis hairy root with theproliferation of multiple proliferation multiple method,and determined flavonoid glycosides ofmedium Vaccaria segetalis and hairy root with HPCL, investigating the effects of exogenoushormones on growth of Vaccaria segetalis hairy root and flavonoid glycosides of mediumVaccaria segetalis, such as2,4-D,6-BA,NAA,KT salicylic acid, methyl jasmonate, yeast extractCa2+,Ag+,the results show that:The type and concentration of hormones and elicitors have different effects on Vaccariasegetalis hairy root, and impact on growth of hairy root and secondary metabolites-vaccaria-were not consistent.2,4-D,6-BA,NAA,KT had different effects on Vaccaria segetalis hairyroot’s growth, with increasing concentrations,2,4-D significantly inhibited the growth of hairyroot.6-BA was slightly inhibited the growth of Vaccaria segetalis hairy root, NAA and KTsignificantly promoted the growth of hairy root, and KT was the strongest. These four hormonescould promote the accumulation of flavonoid glycosides of Vaccaria segetalis hairy root,descending order was KT, NAA,6-BA,2,4-D.Salicylic acid, methyl jasmonate, yeast extract,Ca2+,Ag+induced varying degrees of impact on the accumulation of Vaccaria segetalis hairyroot’s growth and secondary metabolites. Methyl jasmonate,Ca2+,Ag+inhibited the growth ofVaccaria segetalis hairy root, salicylic acid and yeast extract have no effects on hairy root,These five elicitors could significantly promote Vaccaria,20mg/L Salicylic acid,150umoL/Lmethyl jasmonate,20mg/L yeast extract,1.0mmol/L Ca2+,10mmol/L Ag+was optimalconcentration.Response sensitivity of cells to elicitors was different, only a certain period cells couldaccept elicitor signal effectively, and it showed the strongest elicitor activity. In the late growthof hairy root, adding elicitors could get maximum flavonoid glycosides. Induction time also hadsome impact on the hairy root vaccaria. When vaccaria reached the highest total, induction timeof salicylic acid, methyl jasmonate, yeast extract, Ca2+, Ag+were36h,48h,60h,60h,36hrespectively. |
