In this study,The cold-adapted cellulose-decomposing strains were isolated, Afteroptimizing the fermentation conditions were combined, inoculated the composting experiments,in order to study the effects of the cold-adapt complex microbial agents on cattle manurecomposting.(1)7efficient cold-adapted cellulose-decomposing strains was isolated from cattlemanure and Changbai mountain by congo red method and flask fermentation,further4strainsthat have not antagonism were achieved with the antagonism experiment, including3fungi,1bacteria. Morphological observation, physiological and biochemical identification andmolecular biology identification were applied. finally NX5was determined to be the member ofArthrobacter sp,F-3I was Aspergillus versicolor, FF2-2was Aureobasidium pullulans, NF3-2was Botrytis elilipticas.(2)NX5、F-3I、FF2-2、NF3-2were optimized by using single factor method.Determining the effects of the five factors that different carbon sources, nitrogen sources, initialpH, fermentation temperature and fermentation time on producing enzyme of strains. The resultsshowed that the optimum carbon source of four strains were wheat bran, lactose, glucose, wheatbran respectively; The optimum nitrogen source were yeast powder, beef extract, peptene,yeastpowder respectively;The optimum pH were8.0,6.0,6.0and7.0respectively;The optimumfermentation temperature were30℃,23℃,23℃and15℃respectively;The optimumfermentation time were3d,5d,5d and3d respectively. FPA and CMCA production volumeincreased to30.25U/mL and260.56U/mL、31.57U/mL and240.33U/mL、34.54U/mL and98.76U/mL、49.72U/mL and356.64U/mL after optimization.(3)Combined the cold-adapted cellulose-decomposing strains with Bacillus subtilis strains.Based on cow dung and straw as raw materials below0℃, seting up three group: inoculatingcold complex microbial agents group, inoculating common microbial agents group and naturalcompost group. Determined the variety of tempreture、pH、C/N、total Phosphor、NH4+-N、NO3--N and GI during the composting process. The results showed that inoculating coldcomplex microbial agents group get to high-temperature phase (>50℃) at the11th day, earlierthan inoculating common microbial agents group, the highest tempreture was62℃,maintained9days, the natural compost group hasn’t reach to high-temperature phase until the end. At the endof the composting, Adding the cold complex microbial agents group and the common microbial (4)agents group’s pH was7.17and7.24Respectively.;the C/N was16.10and15.67respectively.;there were no significant differences in total Phosphor content, the GI was92%and85%, Respectively;Adding microbial agents groups NH4+-N was significantly lower than that ofnatural compost group, NO3--N levels increased; Inoculating the cold complex microbial agentswas able to accelerated the process of composting and ensure the compost quality.
