Abstract/Summary:
Because anthurium andraeanum is ideal material of cut flowers, thus to continuously expand the sales market. But the production, due to the spread of bacterial blight, anthurium industry severely damaged. Prevention and control of the disease, there is no effective way, has become one of the restricting factors of anthurium production development.The occurrence of diseases is the result of the interaction between plants and pathogenic microorganisms, from the perspective of pathogenic microorganisms to elucidate its pathogenic mechanism will provide the effective prevention and control of the disease.Studied in this paper from the perspective of pathogenic microorganisms, by organizing small chromatography separation and identification of the disease pathogen, through morphology, biochemical index, molecular characteristics identification of preliminary identification of the original small kind of types of the disease, and through high-throughput sequencing methods genome, a survey was carried out on the pathogenic bacteria through bioinformatics analysis to find the pathogenic genes, in order to further develop anthurium and bacterial leaf blight pathogen interactions.The main experimental results are as follows:1. Selection of five strains R-X, D, J, R8, R9of Anthurium isolated strains is from diseased leaves of Danzhou.2. Through experiments tieback, we can determine the five isolated pathogenic strains is significantly. The five isolated strains using only the maltose, without using the rest of the fructose, glucose, sucrose, lactose, and glycerin five kinds of sugar. Various strains had no gas phenomenon after metabolism of sugar in vitro duchenne small tube. Five strains can consume the starch, can produce protease decomposition of collagen in gelatin medium, can decomposition cysteine generate hydrogen sulfide in the culture medium.3. J, R8two strains were the results on NCBI blast analysis results show that the two strains with Xanthomonas is a homology of99%bacterium, basically can determine J, the R8two strains for the Xanthomonas.4. Through the whole genome sequencing Anthurium pathogen, translate R8in prediction of protein in each database and compared them to other Xanthomonas is a bacteria bacterium, find the same and different domain structure, further forecast the R8strains belonged to Xanthomonas axonopodis disease-causing genes. General gram-negative plant pathogenic bacteria have Hrp (hypersensitive reaction and pathogenicity gene) and Avr (avirulence gene) two systems to regulate the interactions with plants. Through the comparison of the database to find out for measuring strain with disease-causing genes in the genome,there are38Hrp genes,and30Avr genes, Through the analysis to find these genes is helpful to the future anthurium bacterial leaf blight pathogen research and anthurium disease pathogenesis research.
