Pumpkin is one of the most widely cultivated vegetable in China, it had many various fruit color, high nutrition with high economic value. Molecular marker,which could detect the genetic diversity at DNA level, is a powerful tool to study crop gene mapping and construction of linkage map. In this study, two high-generation inbred Cucurbita maxima (06820-1) as male parent was flesh yellow, orange rind, flesh white, peel gray98-2-351as female, hybridization then a single F1was self-pollinated to produce F2segregating population. Combine SSR,AFLP, RAPD markers and BSA with the early gene positioning of pumpkin peel flesh color, and construct genetic linkage map of pumpkin, The results were as follows:1^Using SSR, AFLP and RAPD, three kinds of molecular markers were used to construct a genetic map, which contained20linkage groups and81markers, included57SSRs,21AFLPs,3RAPDs, the total length of the linkage group was991.5cM, and average distance was12.2cM. The peel orange color was completely dominant of quality traits, and determined by one pair of completely dominant gene. Peel color gene was located in LG5, and the most near loci was PU078072, the linkage distance was5.9cM.2> About peel color, for parents, F1and F2plants to investigate and statistical analysis. The results showed that peel color was controled by eel1nucleus,in the mature fruit, appear orange peel and gray peel segregation ratio by Chi-square verified substantially was3:1segregation ratio in the F2population, according to mendel’s law. The peel orange color was completely dominant of quality traits, and determined by one pair of completely dominant gene. Peel color gene was located in LG5, and the most near loci was PU078072, the linkage distance was5.9cM.3, According to five grade standard for flesh color, for parents, F1and F2plants were analyzed, the results showed:Yellow flesh and white flesh hybrid materials, regardless of orthogonal or reciprocal cross F1were yellow flesh, description yellow flesh was completely dominant trait. Appear yellow flesh and flesh white segregation ratio by Chi-square verified substantially was3:1segregation ratio in the F2population, according to mendel’s law. From the genetic expression show, Ripe pumpkin flesh colors yellow flesh trait was determined by one pair of completely dominant gene.4-. Combine SSR markers and BSA, found a gene closely linked with the flesh color of SSR markers PU132712, by plant verification of F2segregating population, Appear yellow flesh and flesh white segregation ratio by Chi-square verified substantially was3:1segregation ratio in the F2population, according to mendel’s law.
