Sijunzi decoction is the basic formula to strengthen the spleen and its"qi", which is used in the treatment with peptic ulcers, chronic gastritis, chronic colitis, chronic hepatitis, and nearly40kinds of patients with symptoms of "poor spleen". The experiment tries to study the interventional effect of Sijunzi decoction on the acute alcoholic liver injury in mice and the underlying mechanism, including the following four experiments.Testl The interventional effect of Sijunzi decoction on the acute alcoholic liver injury in mice.To investigate the interventional effect of Sijunzi decoction on the acute alcoholic liver injury in mice, we divided100male ICR mice into control group, acute alcoholic liver injury model group,Sijunzitang high, medium and low dose of Sijunzi decoction groups (20,12,4g/Kg) randomly. The mouse was orally administered the corresponding doses of Sijunzitang once a day.After12h, alcohol was given about5g/Kg B.W. every day for consecutive18days (approximately three courses).5mice of each group were sacrificed to compare the liver index, serum ALT, liver pathology At D2, D7and D19. The results show that, compared with the model group, mortality was significantly reduced in the low, medium and high doses of Sijunzi decoction. At D2,the lesions of the model group were similar to the other drug groups,however the high dose group was a little better.At D7, the tissue lesion of model group was severe,the liver index decreased significantly, ALT increased, but low,middle and high Sijunzi decoction group showed better therapeutic effect. Morever,low, middle, high doses of Sijunzi Decoction alleviated the pathological changes in the liver of mice at D19.Test2Study the impact of the Sijunzi Decoction on the subcellular structures of acute alcoholic liver of miceTo observe the effect of the Sijunzi Decoction on the subcellular structures of acute alcoholic liver, we selected the livers of the most severe lesions of the model group at D7as the sampling to conduct TEM observation. The model group showed irregular cells, concentrated nucleus, mitochondrial swelling,some crushed mitochondrial and a large cytoplasmic vacuoles. The nucleus of the low,middle and high groups are round.The mitochondria were more normal compared with the other groups. At the same time,the area and the mount of the cytoplasmic vacuoles were reduced significantly with the increasement of the dose.Test3The antioxidant effects of Sijunzi decoction on acute alcoholic liver injury in miceIntaking excessive alcohol may cause great oxidative stress that results in lipid peroxidation, reducing mitochondrial GSH and methionine,which eventually injurying to the liver cells. To assess Sijunzi decoction on acute alcoholic liver injury in mice, we detected the activities of T-SOD and GSH-PX, content of MDA and the expression of CYP2E1of each group.The results showed that Sijunzi decoction didn’t improve T-SOD activity, but high doses (20g/Kg) of Sijunzi decoction improved GSH-PX activity of acute alcoholic liver injury in mice, middle (12g/Kg) to high (20g/Kg) dose of alcohol can reduced MDA content of liver homogenate and the expression of CYP2E1of the acute liver tissues, low dose (4g/Kg) decreased the expression of CYP2E1until D7.Test4The impact of the Sijunzi Decoction on the spleen lymphocyte proliferation Mesure the effects of different concentrations of Sijunzitang decoction on lymphocyte proliferation to provide evidence of " tonifying spleen" of Sijunzitang decoction at the cellular level. The cells cultured in the cell medium added Sijunzi decoction with ConA for44h,68h. Add MTT4hours later,and then detect OD values. The results showed that cell proliferation was strengthened combined with ConA compareing with Sijunzi decoction alone added. Exceeded2μg/mL,Sijunzi decoction inhibited cell proliferation, while promoting effect of less than0.5μg/mL of Sijunzitang decoction little.Only within the concentration range of2-0.5μg/mL, Sijunzi decoction promoted cell proliferation significantly.
