The quantity and quality of cashmere were influenced directly by the trait of skin follicle. In this study,on the basis of hair follicle growth and development in the cashmere goat,researching the differences in the expression of microRNA and genes between skin of fetal and lamb, to clarify the cashmere goat skin hair follicle growth process of how microRNA regulate gene expression. The results are as follows:1. Construction of a70-day fetal skin small molecule RNA library,192cloned plasmid DNA be sequence. Screen and compare in GenBank and mirBase by Blastn, find that32sequences are highly homologous to other animal miRNAs. These miRNAs classify into19different miRNAs, and there are nine have registered in GenBank for adult sheep cashmere goat skin miRNA, and the remaining10are the new cashmere goats miRNAs.2. The differentially expressed miRNAs are screening between skin of70d fetal and two weeks lamb by Exiqon miRNA8.1expression profile chip. There are4miRNAs up-regulated in Cashmere goats70days fetal skin compared with lambs. These up-regulated more than five times the miRNA genes for miR-376a, miR-451, the other two are miR-487b and miR-487b, respectively, up-regulated3.32and2.46times.64miRNAs are down-regulated in70days fetal skin compared with lambs, and10are, respectively miR-145、miR-270、miR-101_MM1、 miR-31、miR-21_MM1、miR-200a、 miR-165a/、miRPlus17860、miR-202*、iR-9b*。...
