Cashmere goat is an excellent sort of goat used for hair, which growth of skin and hair is regulated by many kinds of factors. It has been confirmed that melatonin can regulate the growth of the goats’ hair follicle on skin, moreover many genes and miRNAs all involve in the growing development of the hair follicle on skin. Although there are lots of researches on the development of the hair follicle on skin, no clear method is carried on to filter the relative genes. This experiment is conducted to verify the feasibility of the method which start to begin with melatonin and miRNA in order to filter the relative genes, and detect the expression of these relative genes, at last some important genes,such as FGF5、KRTAP and GSDMA were filtered.The following conclusions were obtained from the study.1、Establishing a sample bank of down producing goats’skin, hair and cashmere which information content is complete.2、The method using the melatonin and MTFP to filter the genes related to the development of the hair follicle on skin is feasible, and the operation is simple and convenient. Some important genes,such as FGF5、KRTAP and GSDMA were filtered.3、Heeling-in melatonin significantly upregulate the expression of FGF5in cashmere goat skin, strengthen the relationship between the FGF5and β-catenin,NTRK3,hairless,etc genes, the effect of melatonin on complex tissues and single cell is different.4、The role of FGF5gene in goat hair phenotype is different from other animals; MAPK signaling pathway is the greatest impacted by melatonin.5、The seven miRNAs which we selected are all involved in the cyclical development of skin and hair. The expression level of descending order is miR-720,miR-205,miR-203,let7a,miR-199b,miR-200b and miR-25. the expression levels of every miRNA were significantly associated (R>0.91, P<0.0001);It’s negative correlation between miRNA expression level and number of target genes, the correlation coefficient was-0.43.6、The seven miRNA played a important role in material transportation, shin and hair structure, Gene imprinting, Precursor mRNA splicing, Cell apoptosis, DNA methylation, Transcription and extends, Cell structure. Thus regulated the hair follicle development of cashmere goat. In addition, we also made the following assumptions after analysis:melatonin can upregulate the expression of FGF5by regulating the RORa in cashmere goat skin, FGF5can promote hair growth by positively regulatingp-catenin, NTRK3, hairless and Wnt10b,inhibiting the function of MSX2and BMP2.In summary, this study provided a physical and theoretical basis for the future research of cashmere goat, and also provided a feasible methods and ideas for future research...
