P5CS transgenic plants and their progeny obtained before were used as materials, and the genetic stability of exogenous gene expression in different generations plants of T0, T1and T2transgenic wheatgrass were determined by molecular detection in this study, which would provide theoretical basis for the research and application of transgenic wheatgrass; at the same time. P5CS of T0, T1and T2generation transgenic wheatgrasses were trained by salt stress, and the salt resistance were determined. The aim was to provide the theoretical basis for the creation of new resistant germplasm, to breed the new varieties of transgenic wheatgrasses. The results of this study were as follows:(1) PCR analysis, dot blot and Southern blot results showed that the Genome of T0, T1and T2transgenic wheatgrasses contained P5CS gene, the P5CS gene integration number were1-2in three generation of transgenic wheatgrasses. Detection the P5CS gene of transformation rate is31.82percent in T0generation plants and that are26.3and29.17percent in progeny (T1and T2) generation plants respectively. All of above proved that the P5CS gene in transgenic wheatgrasses could be inherited stably.(2) Under the salt stress, compared with the control plants, leaf relative water content (RWC) and the soluble protein content of T0, T1and T2transgenic wheatgrasses decreased gradually, but the content of Pro and membrane permeability increased, and the content of chlorophyll decreased first and then increased, the content of MDA increased in the first week and then decreased in second week;at the same time, the changes magnitude of RWC, Pro, chlorophyll and soluble protein in T0. T1and T2transgenic wheatgrasses were always higher than the control plants, but membrane permeability and MDA were the opposite. In the antioxidant system, the activity of SOD, POD, CAT increased first and then decreased, and were higher than the control, but between the T0, T1and T2transgenic wheatgrasses were no significantly different. Which suggested that the T0, T1and T2transgenic wheatgrasses; salt tolerance inhanced obviously, and the P5CS gene could be stably inherited and expressed between T0, T1and T2generation.
