Cordyceps sinensis, one of the famous traditional chinese medicinal material in China, is considered as one of the admiring health prodμcts in modern times. It is also a hot issue in modern traditional chinese research. However, the resource of Cordyceps sinensis is reducing gradually. Aiming at extending a substitμte for Cordyceps sinensis and providing experience for clinical medication and People’s daily use, detailed stμdies focusing on identification method and chemical composition were carry out on artificial cultivation of fruiting bodies of Cordyceps sinensis in this paper.In order to ensure the purity of the product, water, total ash and extract of artificial cultivation of fruiting bodies of Cordyceps sinensis were determined in this research.The TLC method was employed to confirm the medicinal ingredients and medicinal value of the artificial cultivation.The results showed that the artificial cultivation contained cordycepin and adenosine.The above results presented preliminary proof of the possibility that artificial cultivation of cordyceps fruiting body may substitμte cordyceps sinensis.The cordycepin and adenosine were determined simultaneoμsly by HPLC. The C18(250mm×4.6mm,5μm) column was used with mobile phase consisting of methanol-water(20:80); The detection wavelength was set at260nm and the column temperature was30℃, the injection volume was10μl. In this condition, good resolution of cordycepin and adenosine was achieved. The content of both cordycepin and adenosine was more than0.08%, which was much higher than that stipulated in the Chinese pharmacopoeia(not be less than0.01%).Sinensis polysaccharide was determined by phenol-sulfμric acid method, the result showed that sinensis polysaccharide in artificial cultivation of cordyceps fruiting body was more than0.10%.Sodium thiosulfate titration method was used to determine the content of mannitol in the artificial cultivation of cordyceps fruiting body, the research results showed that the content of mannitol was not less than4.8%.
