A Study Of Polyamine Metabolism Related Mechanisms In Peach Bud Dormancy Process

The reseach had been carried out in horticultural experimental station and laboratory of protected pomology of Shandong Agricutural University from2011to2014with ’Shuguang’peach as the experimental material, and a series of studies as follows:(1) Changes in antioxidant enzyme activity, antioxidant content, H2O2content during dormancy in flower bud of Shuguang nectarine;(2) Changes in polyamine species,content, arginine content and related polyamin metabolism enzyme activity during dormancy in flower bud of Shuguang nectarine;(3)After dormancy breaking treatment, changes of the nectarine flower bud germination rate,and the related enzyme activity.The main results were as follows:(1)It can be seen through the determination of the enzyme activity of CAT, SOD, POD and the MDA, the soluble protein content during dormancy in flower bud of Shuguang nectarine:the CAT activity increased in early fall dormancy, maitain a low level during the deep dormancy period and increased again after that;the activity of POD increased and maintained at a high level in the early dormancy period, reduced in the deep dormancy and increased slightly in the dormancy breaking period;there are some similar changes of SOD and CAT activity, slowly incread in the early dormancy period and then decreased, increased in the late dormancy and decreased again;the MDA content continued increased in the early dormancy and maintained a low level until the deep dormancy;the soluble protein content remained relatively stable state,and reached the highest peak of themselves respectively in the early and the deep dormancy.The content of H2O2greatly rised in deep dormancy and decline after reached the highest peak in the dormancy breaking period.(2)It can be get through the determination of the polyamine content during dormancy in flower bud of Shuguang nectarine:the highest content is Put, next is Spm,and the lowest is Spd; the Put content increased as the satisfied of the chilling the Shuguang nectarine got, decreased in the dormancy breaking period; Spd and Spm contents maintained an upward trend in the entire dormancy process, and rised significantly in the dormancy breaking period. The content of arginine had a little changes and maintained a stable, it increased greatly in the dormancy breaking periodlt can be get through the determination of the polyamine metabolism related enzyme activity:there is little changes of ADC activity in the early dormancy period, the ADC activity decreased at first and then rised to the highest peak in the deep dormancy period, it would decreased again after the peak;the activity of ODC relatively stable in the early dormancy and decreased in the dormancy breaking period;the activity of PAO is much higher than the DAO activity, their changing tendency is similar that elevated in the dormancy period and decreased after reached the highest peak in deep dormancy period.(3)For dormancy breaking treatment of Shuguang nectarine buds that:the flower buds’ germination rate have all reached more than40%after high temperature,75mg/LTDZ,0.5%HC processing;after the dormancy breaking treatment, ADC activity increased slightly and then continuously decrease, the ADC activity is higher after high temperature than the other two treatments; the activity of ODC after the treatment reached the48h’highest value decreased slightly, the change of DAO activity is similar to that of ODC, decreased after the highest value in48-96h, in which DAO activity bud was continuously decreased after0.5%HC treatment;the activity of PAO was stable, increased at first and then decreased after high temperature treatment, the other two treatments showed a rising trend.
