| Spider Dragline Silk is a kind of natural protein fiber which is synthesized by a pair of main ampullary gland in spider, It is known as "bio-steel" and has broad application prospects. In order to improve the quality of sheep wool effectively. It is feasible to express spider silk protein gene in wool of Xinjiang Merino sheep by somatic cell nuclear transfer technology, our study is of great significance to improve the commercial value of fine-wool sheep wool fibers.In this study, we made use of Merino sheep fetal fibroblasts as nuclear donors, enucleated oocytes as recipients, prepared716Transgenic Reconstructed embryos by somatic cell nuclear transfer technology,then we took part of reconstructed embryos which had developed to mulberry period for testing by RT-PCR,the positive recombinant cloned embryos were transfered into44recipients,12of which were pregnant, we got7offspring finally.After detecting the transgenic Spider Dragline Silk Protein Gene wool sheep at the DNA, RNA levels, the results indicated that the gene was integrated into the host genome and the gene expressed successfully; Then we tested the toughness of transgenic and non-transgenic merino sheep wool fiber, the results manifested that tension and elongation of transgenic merino sheep wool are higher than non-transgenic fine wool. |
