The wool samples of Xinji and Dongbei fine wool sheep were tested by OFDA2000. The testing results were analyzed by SPSS11.5. The results show: 1.the mean wool fibre diameter of Xinji and Dongbei are (20.28±1.72)μ m and (20.29± 1.77) μ m respectively, the wool fibre diameter of Xinji is finer than Dongbei, but not significantly (P>0.05) ;2.the mean wool fibre length of Xinji(12.32± 1.75)cm is significantly longer than that of Dongbei(11.29± 1.82)cm(P< 0.01 ) ;3.the mean natural wool fibre length of Xinji(8.12±1.39)cm is also significantly longer than that of Dongbei (6.78±1.96)cm(P<0.01 ) ; 4.the fercent yield of clean wool of Xinji and Dongbei are (57.58±6.79)% and (48.65 ± 11.22)%respectively, the difference is significant(P <0.01 ) ;5.the mean wool curves number per centimeter of both flocks are more than 4,the difference is not significant (P>0.05) ;6.there is high correlation between yield of wool and mean fiber length, mean fiber length and natural mean fibre length (P<0.01 ) .To further study the genetic characteristics in two fine wool sheep flocks, DNA samples were obtained from blood of 20 Xinji and Dongbei fine wool sheep and amplified with 40 random PCR primers. The polymorphic makers were analyzed by SPSS11.5. The results show, all of the 12 primers selected can generate polymorphic bands, and the length of these DNA fragments is between 300-3000bp.The common bands frequency in the two flocks is different. We found a unique band(973 bp ) of f21326 in Dongbei flock which is not detected in other flocks. The genetic similarity among Dongbei flock is higher than that of Xinji flock. The genetic distance between the two flocks is smaller than former reports. ANOVA and T-test show that several RAPD markers are associated with wool traits. The genome DNA polymorphic frequency of Dongbei was 52.08%,the polymorphic makers f 21326A, f 21327F, g 2739C, f21328E have significant (P <0.05 or P<0.01) effect on wool traits and body weight. The genome DNA polymorphic frequency of Xinji was 54.32%,the polymorphic makers f21344A, f21326G, f21339C, g2742C have significant (P<0.05 or P<0.01) effect on wool traits and body weight.
