Rumen cellulolytic bacteria strains were isolated from sheep using Hungate roll tube method. They were identified through bacterial physiological and biochemical characterization and16s rDNA sequence homology analysis. The carbon source degradation rate, gas production amount, cellulase activity, fermentation parameter and other indicators of three strains were studied under different cellulose ratio.1. Four rumen cellulolytic bacteria strains were isolated from sheep. They were identified as Clostridium beijerinckii, Clostridium butyricum, Ruminococcus albus, Enterococcus faecium.2. The cellulose-degrading characteristics of Clostridium beijerinckii, Ruminococcus albus, Enterococcus faecium were studied in this study. The carbon source degradation rate, gas production amount, bacteria protein production, cellulase activity, acetic acid content of the three strains had a tendency to increase when the cellulose ratio decreased from10:0to7:3and5:5. The pH had an opposite tendency. The concentration of NH3-N was greater in the cellulose ratio of10:0.3. Ruminococcus albus had the greatest cellulase activity among the three strains. The FPAase, CMCase, microcrystalline cellulose enzyme activity,β-glucosidase, pectic enzyme activity of Clostridium beijerinckii was higher than Enterococcus faecium. The Xylanase of Enterococcus faecium was higher than Clostridium beijerinckii.
