Firstly, we used cellulose(papermfilter) as the only carbon source, four kinds of bacterium (These new species are tentatively name as bacillus H1.H2.H3.H4.)that is highly cellulolytic in the rumen of sheep fed high-fiber diets are isolated on the selected medium. They are identified.These are C.cellobioarum,R.flavefaciens,R.albus,F.succinogenes according to its morphological characteristics and culture characteristics.Secondly,according to the celB gene sequence registered in GenBank,we designed one pairs of special primers for the celB gene. By primer polymerase chain reaction (PCR),the DNA fragment about 407bp was amplified from R.albus,furthermore,the fragment was inserted into pMD19-T vector and identified with PCR,the positive recombinant clones was sequenced and analysed.it was the celB sequence registered in GenBank.The results suggested that celB gene was cloned successfully and all should provide effective experimental materials to further research cellulase.
