Apolipoprotein A5(ApoA5) is a new member of the apolipoprotein family. It was found in2001and played an important regulatory role in TG metabolic, Overexpressed of ApoA5had significantly reduced the level of TG. The mechanism of ApoA5impacts on lipid metabolism have become a hot point in recent years, but there is less reported about the association of polymorphism with the growth-related traits and the developmental expression of triglyceride metabolism-related genes.PCR-SSCP was used to analyze single nucleotide polymorphism of the5’regulatory region of ApoA5gene in461samples from eight different groups. The results indicated that the products amplified by primer P2、P6、P8、P9and P11displayed polymorphism, and there were significant differences in allele frequencies among pigs. Subsequent analysis of genetic diversity proved that moderate and low polymorphism existed between populations, as well as genetic differentiation. The main variation derived from the discrepancy of individuals within populations (75.02%). The results of associated analysis showed ApoA5gene in pigs have a higher genetic diversity.Twelve single nucleotide polymorphisms were detected by sequencing different genotypes in the5’regulatory region of ApoA5gene. According to the Linkage Disequilibrium Test, we detected7tag SNPs, and built11haplotypes. The result of the association of the haplotypes with the traits showed the weight and body size of Hap6(CTGATCC) samples were higher than other haplotypes.These results suggest genetic variants of ApoAS are associated with the growth and developmen of Shanxi white pigs, so ApoA5was consider as a marker for the research of growth in pigs and other livestock.To investigate the developmental expression patterns of ApoA5, ApoC3, PPARα and LPL (four TG metabolism-related genes) in the liver tissue of Mashen pigs and Large White pigs by real-time fluorescence quantitative PCR. The results showed that the expression level of ApoA5、ApoC3、PPARa. and LPL mRNA in liver tissue of Mashen pigs was higher than that of Large White pigs.At the same time, the developmental expression level of each gene was significantly different between species and between ages (P<0.05). The evidence suggests the expression of TG metabolism-related genes in liver tissue of pigs plays an important role on the regulation of body fat deposition...
