Years of traditional vegetative propagation of the local water chestnut (Eleocharis ducis) cultivar"diantou sanwang" has induced the degradation of cultivar characters has been an increasing problem and the fast propagation of tissue culture, acclimation and cultivation technique is the best way to solve the problem. Here, based on the technique system of tissue culture, a field test were done to reveal the standardized cultivation techniques, including seedlings comparative field test, transplanting density and fertilizer test of test-tube plantlet, base fertilizer comparative field test, study on shoot cutting propagation technique and control of stem blight trial. Meanwhile, A convenient and practical fast propagation and cultivation technique of water chestnut are proposed for the local production of water chestnut. The results were as follows:1. The rapid propagation system of local water chestnut was established. The optimal induction medium and proliferation medium were MS+BA1.5mg/L and MS+6-BA1.5mg/L+NAA0.01mg/L+PVP400mg/L, respectively. And the highest proliferation rate reached13.85. The optimal combination of rooting medium was modified as1/2MS+NAA0.05mg/L+AC1000mg/L or MS+NAA0.05mg/L+AC1000mg/L, which rooting rate reached100%。2. The suitable reagent for controlling browning in secondary culture is PVP. Adding400m/L PVP into the medium decreased effectively the browning rate from89.6%to2.0%and increased the multiplication coefficient from11.2to13.85.3. Survival rate of the test-tube plantlet after rooting could reach100%.4. We summarized and generalized the shoot cutting propagation technique.5. We proved that animal manure was not suitable in water chestnut field planting.6. Based on controlling of stem blight trial, we suggested that the chemical "Jiukang Meisu" should not be used on prevention of the stem blight during water chestnuts planting.7. A standardized cultivation technique was established in the test-tube plantlet of water chestnuts:Intensive cultivation; No organic fertilizer; Test-tube plantlet should be taken care before transplantation; A small amount of fertilizer irrigation should be done during the semi-late period; Be sure to do the transplantation in good time; Green manure is better than animal manure as base fertilizer; Right amount of after manuring should be done at the right time; Fertilization should be finished before Bailu. Irrigation should consist in the entire growth period and stop25days before harvest."Jiukang Meisu" should not be utilized to control the stem blight of water chestnuts.
