Notholin hyacinthinum, a kind of perennial herb which belongs to the Liliaceae, hasmedical effects on easing pain and relieving cough, and has special effect on the treatment ofliver cancer, stomach cancer, esophageal cancer and other diseases when coordinated withother drugs. However Notholin hyacinthinum nearly depleted because it grew in highmountains, natural populations were the main source for the plants, it had low naturalreproductive rate, and there was a long process of over exploitation. A rapid propagationsystem through plant tissue culture techniques was established by using Notholinhyacinthinum as material. Based on the investigation of effect of types of explants and plantgrowth regulators on callus induction, the effect of plant growth regulators on adventitiousshoot formation and rooting, and so on, the main findings are as follows:1. The vanes, bulb scales, scale leaves, small bulbs and adventitious shoots of Notholinhyacinthinum could all be used as explants, and the highest callus induction rate was obtainedwhen using scale leaves as explants.2. The highest induction rate of90.5%was achieved on MS medium supplemented with0.5mg·L-1NAA when using scale leaves as explants. Medium supplemented with0.1mg.L-1NAA was more suitable for callus proliferation.3. The highest induction rate was achieved on MS medium supplemented with0.1mg·L-1TDZ for adventitious shoots differentiation. But the optimum medium was MSmedium supplemented with1.0mg.L-16-BA and0.1mg.L-1NAA.4. The optimal medium for rooting was1/2MS medium supplemented with1.0mg·L-1NAA, the induction rate was91.4%.5. When small bulbs were used as explants, embryogenic callus induction rate washighest on MS medium supplemented with TDZ and NAA, which could reach95.6%.6. After being transferred to the medium of lower concentration of2,4-D (0.1mg·L-1), theembryogenic callus differentiated into the multicellular proembryo. On the medium withoutplant growth regulator, globular embryo, elongated embryo and cotyledon embryo wereobserved. 7. Embryogenic callus and non-embryogenic callus were distinguished throughhistologic observation, and the development of the somatic embryo was revealed as well.
