To use male sterility for producing F1hybrid seed is a major approach to usetheheterosisof rapeseed. Thermo-sensitive genic male sterility as one of most importantmethods to make use of heterosis after cytoplasmic male sterility and genic male sterility, ithas not only a wide spectrum of restores and great potential in hybrid seed production, butalso can simplify the procedure and decrease the costing of hybrid seed production as itcan beused for both male sterile line and maintainer line. Now, thermo and/or photoperiod sensitivemale sterility have made preliminary success of hybrid seed production in rapeseed, and thesuccessful popularization of two-line hybrid Brassica napus varieties such as Xiangzayou-5and Ganliang you-2et al. proving it has a great potential in the crops production. However,the basic researches of it especially in developmental aberrations leading to male sterilityhave lagged behind a lot, and this will extremely restrict the utilize of male sterile line as wellas the development of two-line hybrid rapeseed.Thermo-sensitive genic male sterile (TGMS) line Huiyou50S was derived from aspontaneous semi-sterile plant which was found in Brassica napus cultivar ofHuiyou50, andthrough many years of systematic selection. In this study, to make further analysis of themechanism of inheritance and fertility transition of Huiyou50S, artificial climate chambertestand screening of SSR molecular markers were conducted. Meanwhile, it could evaluatethe value of male sterile line by investigated the temperature reaction of Huiyou50S, andmake foundation of the fine location even cloning of the male sterility gene by means ofscreening molecular markers to find the linkage group of male sterility gene of Huiyou50S.The main results are as follows:I. In artificial climate chambers test, five different temperature conditions at oneillumination condition were set and the result showedHuiyou50S was a reversethermo-sensitive genic male sterile line, it was malesterile when at low temperature condition,but fertile under high temperature condition, the stagnation temperature was closed to22℃(day)/12℃(night).II. In the screening of SSR molecular markers, the F2of Huiyou50S an Zhongshuang11 were used for research materials, conbine Bulked segregation analysis (BSA) andBulked-extrems and recessive-class approaches (BERCA). And we have found8SSRmolecular markers linked with the male sterility gene of Huiyou50S, which were alllocatedon A10linkage group of Brassica napus genome. Four of them are not very tightlylinked with male sterility gene and other of them are closed with male sterility gene relatively,the closest markers of BrGMS287was6.00cM genetic distance to male sterilty gene ofHuiyou50S.
