Lagerstroemia indica is one kind of garden ornamental plant with unique appreciating value andgraceful appearance. Data display：Current researches on Lagerstroemia indica mainly focus on its cultivation,different types and garden plant pests control etc. Its reproductive modes contain sowing, cutting and so forth.Some improvements have been achieved in radiation breeding, crossbreeding and tissue culture. However,there are poor phenomenons after the seeding process, such as poor growth, the high mortality of seed andshort storage of good seeds. This essay aims at the study of skills of indoor tissue culture with study results asfollows:1. The choice of sterilization time and browning prevention:(1)Use0.1%HgCl2，the best sterilization time forstem-tip and blade is4min,for stem is8min. The best sterilization time for the axillary buds cut in autumn is10min, which is longer than the ones cut in spring since the pollution rate is high in autumn.(2) The size,physiological state, drawing season and antibrowning has certain influence on the browning and induction, i.e..2~3cm blade is beneficial to improve the induction rate and reduce the browning rate. May and June are thebest seasons for the tissue culture of Lagerstroemia indica. Browning rate is low46.67%and germination rateis86.67%.0.15g/L concentration of activated carbon,0.1g/L PVP and0.25g/LVc has a positive effect onbrowning prevention and activated carbon is better than PVP and Vc. Dark treatment for10d can reducebrowning rate and improve induction rate.2. The rapid propagation cultivation of blade:On account of reducingcost, the best original culture medium ofstem is MS+6-BA1.0mg/L+NAA0.1mg/L and the best subculture is MS+6-BA2.0mg/L+NAA0.3mg/L orMS+6-BA2.0mg/L+NAA0.1mg/L+KT1.0mg/L. The best PH for multiplication culture is5.7and the besttemperature is25~30℃.0.15g/L activated carbon is conducive in improving multiplication ratio to3.67.PVPhas little effect on the multiplication culture. The best rooting medium is1/2MS+IBA0.5mg/L.0.3g/L~0.5g/Lactivated carbon may conduce to the growth of stem.0.1g/L activated carbon can promote the rooting processof tube seedling while high concentration is bad for the rooting.3. The sterile propagation cultivation of Lagerstroemia indica seedling: as for the seeds of different maturities,the sterilization time is also different. It is better to sterilize capsule for8min while the seed for10min. Withthe same sterilization time, the germination rate capsule is48%, indehiscent capsule22%, dehiscent seed19%,seeds stored for a year2%, which indicates that the influence of episperm has on germination is greater thanthat the different maturities of the seeds have. Light has stimulative effect on the germination ofLagerstroemia indica seed. Hormone is conducive in the germination. MS+6-BA1.0mg/L+NAA0.3mg/Lpromotes the highest germina-tion rate. The best multiplication culture medium is MS+6-BA1.0mg/L+NAA0.1mg/L++KT1.0mg/L, the best rooting medium is1/2MS+IBA0.5mg/L. 4. Sterile culture of hydroponic seedling: the activity of seed will descend after storing for a certain time.75%alcohol and0.1%HgCl2is not suitable for the sterile culture of hydroponic seedling. HgCl2may result inserious mortality of hydroponic seedling.5. Sterile culture of hardwood hydroponic buds:Diameter,variety,remove way of leaf biade and hormonetreatment has certain influence on germination speed of hardwood.The germination rate of stem section up to56.67%witch diameter was1.5~2.5cm.The germination speed is as follows: the stem without leaves> stemwith petiole> stem with half leaves. The germination speed of immature stem is faster than hardwood, but thegermination rate and quality is inferior to that of hardwood. Different areas result in different germination rate,Lagerstroemia subcostata Koehne> Lagerstroemia limii Merr> Lagerstroemia indica. The germinationspeed of stem with different concentration of6-BA is faster than that of stems without any hormones. Thegermination rate of20mg/L concentration can be72.27%. Sterilization with mercury chloride for4min is thebest with65%germination tare and28.34%pollution rate. The best multiplication culture medium isMS+6-BA2.0mg/L+NAA0.3mg/L. The best rooting medium is1/2MS+IBA0.5mg/L.6.Acclimatization:Transplanted into pot with the matrix of sand: peat=1:1after culture indoor domesticationthree days and open a bottle of hardening five days, the survival rate was62%.
