This paper studied regeneration system from tissue culture of the soft seed pomegranate which included the key technologies of cultivation on embryo germination, proliferation , seedling strengthening, rooting, refine seedlings and decreasing pollution and browning, multiplication, rooting of explants. Lay the foundation of callus induction, genetic transformation breeding and polyploidy breeding in pomegranate. The results of this study are as follows:1.It is conducive to germination which the seeds were preserved 4 days under 4℃and was cut off part of seed coat, the best medium for germination and subculture was MS + BA 1.0 mg/L + NAA 0.1 mg/L + GA3 2.0 mg/L.2.The best multiplication medium was MS + 6-BA 0.5 mg/L + NAA 0.5 mg/L + GA30.5 mg/L, the influence degree of the plant growth substances on coefficient of proliferation was 6-BA>NAA>GA3.3.The optimum seedling strengthening medium was MS+IBA0.1 mg/L, the plants after strengthening would root shortly.4.The best rooting mediums was 1/4 MS + IBA0.1 mg/L + PP3331.0 mg/L, the rooting rate was 85%, the effect extent of plant growth substances on rooting was IBA>MS>PP333. The best transplanting mediums was soil.5. When treated by 75% ethanol for 30s and followed by 0.1% HgCl2 sodium for 10 min, the explants were kept in culture room for 3d, and then the explants were treated by 0.1% HgCl2 sodium for 10 min again. The success rate was the highest by this method.6.The optimal media for reducing browning rate of the stem segment was MS + 6-BA1.0 mg/L + IAA 0.5 mg/L + PVP 2500 mg/L, the browning rate was 13.6%, the effect of the factors in sequence was PVP>6-BA>MS>IAA.7.The best multiplication medium was MS + 6-BA 0.5 mg/L + NAA 0.1 mg/L, the effect of the factors was 6-BA>GA3>NAA.8.The best rooting medium was 1/2 MS + IBA0.1 mg/L+ PP333 0.5mg/L, the rooting rate was 85%, the effect of the factors was IBA>MS>PP333.
