Qinba Cordyceps militaris(Qinba C.militaris)is the species of entomogenous fungi inthe family Clavicipitaceae. It is naturally distributed in Qinba mountain area of Shaanxi Province. Itcontains many biological bioactive materials, such as cordycepin, polysaccharide, and it hasmany biological activities. For the purpose of obtaining the high cordycepin concentration ofQinba C.militaris regenerated-protoplast strains, protoplast mutagenesis conditions werestudied, and the regenerated progenies were screened. And the biological characteristics of analbino mutant strain(WH-1)from Qinba C.militari(sCM-16)were investigated aditionally.The main test results are as follow.1. The six strains of Qinba C.militaris were cultivated under the same culture conditions.To assess their production performance, the fuzzy comprehensive evaluation method wasused. The results showed that CM-16and WY-1strains were superior strains, and CM-16wasselected as a start strain for mutation.2. For the purpose of studying the optimal conditions of Qinba C.militaris protoplastsisolation and regeneration, the enzymolysis conditions were studied and the results wereoptimized through orthogonal test. The test results showed that to obtain the highestprotoplasts yields, the best condition of protoplasts isolation was:0.6mol/L NaCl, mixedenzyme,6d spawn age of mycelia, under3h,(26±1)℃; to obtain the highest protoplastsregenerated rate, the best condition of protoplasts regeneration was:0.6mol/L mannitol,mixed enzyme,5d spawn age of mycelia, under2h,(26±1)℃.3. The conditions of Qinba C.militaris protoplasts mutagenesis were studied. The testresults showed that the optimal conditions were23.5~31.5s under300mm with50Wultraviolet lamp, and9.6~13.4min with0.6mol/L nitrous acid respectively.4. To select one bacteria strain which sensitve to cordycepin, as an indicator it couldscreen the regenerated-protoplast strains which produce high amount of cordycepin. The biological characteristics and production performance of regenerated strains were studied.The results showed that the bacterial No.34was selected as an indicator and determined asSerratia nematodiphila through16S rDNA analysis. The regenerated strains M4, M9, M30,M32, M41were screened out. M4, M30, M32biological efficiency both reached more than80%,combining their high cordycepin content, they were superior strains and were worth fortherstudying.5. The biological characteristics and production performance of the albino strain WH-1were studied. The results indicated that the temperature, the initial medium pH and myceliabiomass for hyphae growth had no difference between WH-1and CM-16. The cordycepincontent of WH-1stroma was higher, the production period was shorter, and its appearancewas novel, it was worth further studying.
