Baculovirus is used as insecticide that for its host specificity and security toenvironment, etc. Cydia pomonella granulovirus (CypoGV) is one of the majorbiocontrol agent for the codling moth boring the pome in the world. CypoGV is hardto survive in environment for a long time, the effects of biocontrol was decreased forthat. CypoGV seprarated from Xinjiang China in the extreme sunlight and drought,can be ability to adapt to the extreme environment. Studying on how CypoGVimproves the mechanism of UV tolerance that can be not only advantageous to screenfor virulent viruses, but also enrich microbial content for adaptation to extremelyenvironmental mechanism research, as well as provide the theory of microbicpestisides in arid and strong sunlight regions. The realization of mass rearing codlingmoth, the study improved the rearing media based on presvious formula. Research theinfluence of temperature on the codling moth population, when it fed on the optimumformula B. The results are shown:1. granulin fragments separated from5strains CypoGV in west China werecloned by PCR. It showed several difference between5strains CypoGV andCypoGV-m in granulin fragment and amino acid sequence:(1) comtrasting granulin fragment of CypoGV-zy1, CypoGV-ww1,CypoGV-ale1, CypoGV-ale2, CypoGV-ks1with CypoGV-m, there are3,4,2,0,5mutation sites respectively(2) Granulin amino acid fragment of5strains CypoGV compare to CypoGV-m,it showed that a single amino acid exchange leucine121to proline betweenCypoGV-ks1and CypoGV-m, isoleucine172to asparagine between CypoGV-ale1and CypoGV-m.These results demonstrated that granulin of CypoGVs separated from XinjiangChina,have been mutation. Whether there were relations of granulin mutation andvirus ability to adapt to extreme environment, such as intense ultraviolet or longsunlight, it should be further developed. 2. This research aimed at improving the rearing media based on one formulafrom previous research, compared the effects of5diets on the development andreproduction of the codling moth. the laboratory population life table of codling mothat20,24,28and32℃were constructed when it fed on the optimum formula. Theresults were shown:(1) B formula based on wheat starch, soybean, wheat germ and sucrose, byadding a supplementation of yeast extract to the semi-artificial diet, in contrast tooriginal formula, can significantly increase all parameters. The survival rate increasedinto50.2%, pupa weight increased into26.54mg and oviposition per femaleincreased with85.3eggs. The duration of the lava stage decreased with20.2d.(2) Life table showed that the tempture was the key environmental factors whichaffected population growth. Fertility and population trend index rate reached thehighest at24℃with values of62.4and16.4. The highest female proportion was0.55and the intrinsic rate of increase (rm)0.08, both at28℃. It showed that the optimaltemperature of artificial feeding of codling moth is between24~28℃.These results indicated that the codling moths were fed on formula B addingyeast and previous formula, the former was beneficial for population growth. Theoptimal temperture was determined between24~28℃. All above providesignificant technical parameters for rearing a large-scale of codling moths.
