Shaanxi Guanzhong black pig is cultivated specie and awards first GI products from theMinistry of Agriculture. It is an excellent dual-purpose breed of meat and fat. It has strongcharacteristics, such as disease resistance, fertility, resistance to crude feed and high meatquality.In recent decades, due to the impact of foreign pigs on the domestic market, the pigmarket environment is declining, which leads to the many excellent traits separation changes.As a national pig industry technology system-Reproductive Technologies and GuanzhongPig Purification and Rejuvenation of key projects branch, based on this situation, this studyfocuses on the black pig boar semen quality testing and storing at room temperature, whichcould provide a theoretical basis for improving artificial insemination and breeding.To compare the best recipes of dilutions, this study selected five kinds of commonlyused dilutions in pig semen storage at room temperature, and tests their effects onpreservation and select the best formula.Based on this study, we want to study the mostsuitable temperature for preservation, the optimum dilution ratio, dilution PH changes andantibiotic selection. The results are as follows:1. Guanzhong black pig boar semen qualityAfter taking off the black pig boar fresh semen, the test results is as follows: vitality0.85;malformation rate of0.16; PH7.3; density236million/ml; semen volume250ml; colorwhite.2. Guanzhong black pig boar semen preservation under room temperatureAmong the selected five kinds of formulations, saving ability(sperm motility, membraneintegrity, acrosome integrity, malformation rate, effective preservation days) were screened.Results showed that Ⅲhas the effectively preservation of3days. Ⅱ has the effectivelypreservation of2.5days. Ⅳ and Ⅴ have the effectively preservation of2.0days. Thus, thebest dilutions is No. Ⅲ, followed by No.Ⅰ. And No.Ⅱ dilution is slightly better than No.Ⅳand Ⅴ.In short time, there is little significant difference between No.Ⅰand No.Ⅲ in preservation. However, No.Ⅰhas a lower cost, and is more suitable for the short-termpreservation of pig semen in practice.3. The most suitable temperature for preservation of Guanzhong pig semenAmong the10℃,15℃,20℃,25℃four variables set in this test,15℃,20℃groupshave the basically effectively same results, and are better than10℃,25℃groups.10℃group was slightly better than25℃group, however, the difference was not obvious. Thisresult showed that the most suitable temperature for storing Guanzhong black pig boar semenis15℃-20℃.4. The changes of pH in the pig semen dilution in preservation processpH value of semen dilution showing a decline after the first increase in the change, andfinally stabilized. This is due to changes in the metabolism of the sperm cells which wouldproduce acid metabolites, therefore decreases diluents pH. Then dilution buffer began to playa role in making the pH rise, and finally stabilize. This is consistent with domestic relatedreports.5. The most suitable dilution ratio for Guanzhong pig semen preservationAmong the1:1,1:1.5,1:2,1:2.5,1:3five dilution ratio set in the test, the1:1,1:1.5,1:2three ratios showed better than1:2.51:3, but the difference was not significant. For thepractical production cost considerations, the optimal dilution ratio is1:1.6. Research antibiotics addition in Guanzhong pig semen preservationThe results showed that the preservative effect of a mixture of penicillin and gentamicinis better than that penicillin, gentamicin were added alone. However, the difference is notsignificant. When added alone, gentamicin is slightly better than penicillin in preservationprocess, but there is no significant difference.
