In recent years,due to the irrational use of antibiotics,resulting in some bacterial resistance to conventional antibiotics.If you continue to add in extender with conventional antibiotics to save the semen at room temperature,may make the drug resistant bacteria infection by sows.Semen directional to reduce the use of antibiotics,reduce bacterial drug resistance and provide higher biological safety of Guanzhong black pig semen for artificial insemination,this study added a broad-spectrum antibacterial ability of natural cationic in Guanzhong black pig semen antibacterial peptide--e-poly-lysine with semen stored in room temperature.Firstly,this experiment using high-throughput sequencing method to detect the types of bacteria in different time in semen,and through the 16 SrDNA method to identify the isolated part of bacteria in boar semen bacteria;secondly,choose the common bacteria in semen as bacteriostatic test in vitro strain,minimum inhibitory concentration determination;finally,to explore the different concentration of polylysine in Guanzhong black pig semen preserved at ambient temperature,and the influence of 0.16 g/L e-poly-lysine replacing part of gentamicin effect on preservation of semen at room temperature.The main results of this results are as follows:1 Under room temperature for first days in semen bacteria were gram positive bacteria,such as Staphylococcus epidermidis,Bacillus licheniformis and Aerococcus viridans,preserved mainly Gram-negative bacteria fifth days,such as Acinetobacter lwoffi,Achromobacter xylosoxidans and Stenotrophomonas maltophilia.2.The e-poly-lysine of Acinetobacter lwoffi,Staphylococcus epidermidis,Escherichia coli and Aerococcus viridans had inhibitory effect,minimal inhibitory concentration of Staphylococcus aureus and Escherichia coli was 0.02 g/l,Acinetobacter lwoffi and Aerococcus viridans The minimum inhibitory concentration of 0.04 g/l.were added 0.16 g/l polylysine,adding 50% and 25% standard gentamicin(0.25 g/L)of poor Achromobacter xylosoxidans and inhibitory effect of Stenotrophomonas maltophilia;0.16 g/L e-poly-lysine instead of 50% and 75% respectively with gentamycin add the inhibitory effect of these two kinds of bacteria in the difference between the 0.25 g/L gentamicin effect was not significant(P > 0.05).3 In 0.04 to 0.64 in the diluted semen Tim g/L polylysine does not affect the osmotic pressure(P > 0.05),when adding 1.28 g/L polylysine when increase the osmotic pressure(P < 0.05),0.04 ~ 1.28 g/L polylysine could effectively inhibit the growth of bacteria in semen and improve the preservation effect of semen preservation;after three days at room temperature Among them,0.16 g / L ?-polylysine was the best(P <0.05).4 Epsilon poly lysine and gentamicin in dilute solution Tim With the synergistic antibacterial effect,0.16 g/L e-poly-lysine respectively 50% and 75% instead of adding 0.25 g/L gentamicin can obtain similar gentamicin and bacteriostasis effect of semen preservation at ambient temperature(P > 0.05).In summary,the gram negative bacteria in Guanzhong black pig semen is the dominant bacteria,0.04 g/L polylysine effectively inhibit Staphylococcus epidermidis.Escherichia coli,Acinetobacter lwoffi,Aerococcus viridans growth;adding polylysine to Achromobacter xylosoxidans and Stenotrophomonas maltophilia inhibition effect,0.16 g/L polylysine can replace part of gentamicin to suppress this Two kinds of bacteria;0.04 g/L ~ 1.28 g/L polylysine can improve the preservation effect of semen,of which the optimal dosage is 0.16 g/L;in the semen while adding epsilon has a synergistic inhibitory effect of gentamicin and poly lysine.
