Deciduous tree amur cork (Phellodendron amurense Rupr.) as the Chinese endemic species, is one of the important associated tree species in the northeast broad-leaved Korean pine, and the valuable timber species and medicinal plants. The important secondary metabolites of amur cork Berberine, Ajrtohrrizine and Palmatine, play an important role in clearing heat, dampness, purging fire and detoxicating effect. And it is the raw material of many new drugs and specific medicine. We explore the optimal cultivation conditions for amur cork seeding growth and main alkaloids accumulation from the two aspects which are arbuscular mycorrhizal and nutrient elements. To provide certain scientific basis for the resources protection and effective use of amur cork.(1) We screened three kinds of arbuscular mycorrhizal fungi, at the same time, choose three different sources of annual and biennial amur cork seedlings as the research object in the inoculation experiment.Comparing the test results of annual amur cork seedlings among different provenance that jilin, heihe and space breeding, by combinating the different species and different kinds of sources to explore the impact that the arbuscular mycorrhizal fungi on the growth and alkaloid content of amur cork and its difference among different provenances. The results of this experiment show that the growth and alkaloid content of amur cork with arbuscular mycorrhizal fungi are better than the control group more or less. It indicates that mycorrhizal fungi can effectively promote the growth of amur cork and the accumulation of alkaloid content. Overall, it is the Glomus mosseae that affect the content of berberine and jatrorrhizine mostly, and for palmatine it is Glomus intraradices. Also, from the experimental results, we can also see that it is different among provenances that the impact on alkaloid with arbuscular mycorrhizal, and the difference may be related to the provenance.Contrast the data of annual and biennial amur cork seedings belonging to different provenance of jilin hehei and space breeding respectively,mto explore the difference of the influence that arbuscular mycorrhizae on amur cork seedling growth and the main alkaloid accumulation between provenances. The experimental results of biennial amur cork seedings show that the influence of Glomus mosseae is the most significant in the three arbuscular mycorrhizas expect the the outstanding performance that Glomus intraradices on Palmatine in amur cork root bark of the space breeding. The comparison result of annual and biennial amur cork belonging to different provenances shows that the influence on Ajrtohrrizine is different among the three kinds of arbuscular mycorrhizas, however the difference does not exist in the impact Berberine and Palmatine content. (2) In the experiment of affect that Nitrogen concentration on amur cork seedling growth and major alkaloid content, the culture medium used in the experiment based on the Ultrianen nutrient solution in which the nitrogen concentration gradient is different and the concentration of other nutrients elements are equal.Compared three test results, the rule of amur cork with different concentration of nitrogen treatment is consistent, the seedling biomass is falling along with the concentration increase of biomass in nitrogen concentration of1~6mmol/L range, when the concentration of nitrogen reaches8mmol/L, biomass rised rapidly, and it fell rapidly when the nitrogen concentration reached10mmol/L, then it appeared a slow upward trend along with higher biomass nitrogen concentration. The determination results show that, the biomass is maximum in treatment of the50th days. Three processing results all show that the amur cork seedling biomass is the largest when the nitrogen concentration is8mmol/L, it indicates that8mmol/L is the optimum concentration for the growth of amur cork seedling.The experimental results show that the effect is remarkable that the nitrogen concentration on the main alkaloids yield of amur cork seedlings. In the three kinds of alkaloids, it is proportional to the relationship between the alkaloids yield of berberine and palmatine and the biomass roughly. The maximum biomass occurred when the nitrogen concentration is8mmol/L. The proportional relationship between the jatrorrhizine yield and biomass is not obvious.
