Cordyceps (Cordyceps militaris) is not only nutritious, and have many bioactivities, suchas reducing blood sugar, reducing blood pressure, anti-bacterial, anti-inflammatory,anti-tumor and enhancing immunity, etc. It is an ideal health food.Cordyceps contains a variety of active ingredients, like cordycepin, adenosine, SOD,Cordyceps polysaccharide and ergosterol, these active ingredients is an important indicator ofthe level of Cordyceps stroma quality. The methods of Cordyceps culture include liquidfermentation and solid cultivation. The factors that influence the content of the activeingredient are largely studied in liquid fermentation, and solid cultivation is often used on theoptimization of growth conditions. This paper studies the influence of Cordyceps growingmedia, the main environmental factors and changes in the basic process parameters on theproduction of stroma and contents of cordycepin and adenosine. The main findings are asfollows:1. The effects of carbon sources, nitrogen sources, inorganic salts and triacontanol onCordyceps production and contents of cordycepin and adenosine. The results showed that:(1)adding9g/L soluble starch to the media had a significant impact on the content of Cordycepsproduction and contents of cordycepin and adenosine, but the effects were not significant afterglucose and sucrose were added.(2) When adding6g/L peptone as a nitrogen source ingrowing media Cordyceps grew better, and contents of cordycepin and adenosine were higher.(3) Among the tested inorganic salts,1g/L ferrous sulphate significantly promoted theproduction of both Cordyceps and the active ingredients. Furthermore,2g/L triacontanolcould also promote Cordyceps growth and accumulation of the two active ingredients.2. The influence of the light intensity, illumination time, wavelength and initial pH on thecontent of Cordyceps stroma production and effective ingredient contents was studied. Theresults showed that: when the light intensity and illumination time was150lx and8h/d,respectively, higher Cordyceps and active ingredients production could be yielded. Thoughthe cordycepin content in remaining medium is higher, the light intensity and illuminationtime do not make a large amount of stromal adenosine. Blu-ray benefited the growth ofCordyceps and accumulation of the two ingredients, but there is not much difference in the accumulation of adenosine between other lights. And no matter what the light was, theremaining medium almost contained no adenosine. When initial pH of cultivate medium is7.0,the dry weight of stroma and two active ingredients yield of cordycep are highest.3. The effects of medium amount and water ratio of medium on the production ofCordyceps stroma and contents of two active ingredients were further investigated by usingquadratic orthogonal rotation design experiment. The results show that: the medium dosage of350g/box and medium material to water ratio of1:1.5yielded the largest output of Cordycepsstroma and highest production of cordycepin.
