Wechose the easier and cheaper way to produce.And the products are easier to preserve andtransport.The experiment began with the semen collection method improving.We used thetype of open semen collection to collect the midpiece semen which is rich of sperm.Doing inthis way can made the semen frozen more efficiency.In terms of screening the dilution,the experiment consult the recipe of animalsemen dilution.designed10basic dilution recipe and the frozen dilution which added4%，5%，6%，7%，8%glycerinum.According to the motility rate and the body full rate to screenthe recipe.The result showed that the recipe1and the recipe which added the5%glycerinummade the semen preserved well.In terms of making the frozen semen.We cooled the semen step by step.The strawsfrozen in the liquid nitrogen at last.We optimized the precept of centrifugation and thebalance.The standard of screening is the motility rate and the body full rate,too.We chosed thecentrifugation speed was1500r/min. We equilibrate the semen10min in20℃then centrifugethe semen,after that conduct the second dilution in20℃and equilibrate the semen2h in4℃can get the better result.The motibilty rate of the frozen semen frawed is43%.
