Study On Application Of Youxian Duck Semen Collection And Preservation Technology

In order to establish the foundation of the research and application in the field of artificial insemination and semen cryopreservation of Youxian duck fed on net,a test was conducted to study the technology of semen collecting,diluting,refrigeration,freezing and artificial insemination,in which Youxian duck was used as experiment material.The method and the results of the research are as follows:1.Semen collection technologyFrom 43 170-day-old male Youxian ducks,17 able-bodied individuals with strong sexual reflection were selected to be trained for 15 days to establish reflection.12 ducks could be used to artificially collect semen,the success rate of training was 70.59%,while only 3 ducks could be used to artificially collect semen from 26 untrained male ducks,and the rate was 13.04%.The semen amounts of Youxian ducks were 0.09~0.5 mL,and the average amount was about 0.25 mL.The vitality of fresh sperm was 0.6~0.9 and the semen was acidic.The density of sperm was 3.788~5.885×10~9 sperms/mL,and the sperm densities of induced estrus ducks were all higher than that of massaged ducks.According to the mating pattern and penis characteristics,a semen collection cup suitable for Youxian duck was made.And the cup was superior to the existing ones in terms of simplicity and semen collection rate.2.Semen dilution and refrigerationThe semen of ducks were diluted by LR diluents with osmotic pressure of 303,273,215and 180 mOsm,and stored at 6?.The results showed that the osmotic pressure in which the semen vitality was the most stable was 303 mOsm,and the longest survival time in vitro was150 h.3.Semen cryopreservationThe semen from induced estrus ducks was diluted at 1:4,1:6 dilution ratio and cryopreservated.Sperm vitality in the group which semen was diluted at 1:4 dilution ratio was better than that at 1:6,and the sperm was scattered more uniform.The semen from the ducks(between 170~310-day-old)was frozen and then graded with the vitality.According to the rate of sperm with linear motion,the average vitality of the frozen semen was 0.2~0.4,up to 0.5.With the method of deep insemination by opening the anus,frozen semen and fresh semen(with the vitality more than 0.3)were respectively inseminated.The fertilization rate of fresh semen group was 88.17±4.10%,and the one of frozen semen group was85.93±7.00%,both were very close.The fertilization rate reached the advanced level among the same kind of research at home and abroad.
