In this paper,4cultivars, Fengxian Dahongpao, Hancheng Dahongpao, WuduDahongpao, Gou jiao, were taken as rootstocks, and Wu ci jiao was taken as scion. Throughthe survival rate investigation of4different rootstocks and3different grafting methods, theappropriate rootstock and grafting method to Wu ci hua jiao were confirmed and the changesof related physiological indicators in the healing process were studied, which were expectedto provide the theoretical basis and technical guidance for breeding of grafted seedling andcultivation promotion of Wu ci hua jiao.The main findings are as follows:1. The experiment was carried out with4one-year-old cultivars, Fengxian Dahongpao,Hancheng Dahongpao, Wudu Dahongpao, Gou jiao, as rootstocks and one-year-old branchesof Wu ci hua jiao as scion. The result showed that the survival rate of4rootstocks weresignificantly different, among which the survival rate of Hancheng Dahongpao wassignificantly higher than that of Fengxian Dahongpao, Wudu Dahongpao, Gou jiao, so therewas good compatibility between Wu ci hua jiao and Hancheng Dahongpao. HanchengDahongpao is the appropriate rootstock for the graft breeding of Wu ci hua jiao.2. Different garting methods significantly affected the survival rate of Wu ci hua jiaografting. The survival rate of chip budding was significantly higher than that of cut graftingand tongue grafting, so the latter was the appropriate method for Wu ci hua jiao grafting inSpring. In Fengxian County and the region which has simlar climate to Fengxian County,Hancheng Dahongpao and chip budding are respectively the appropriate rootstock and theappropriate method for Wu ci hua jiao grafting in Spring with the survival rate as high as81.11%.3. During the graft union healing, the activities of POD, CAT and PPO, the content ofH2O2, lignin, IAA increased at first and then decreased. They increased significantly andreach max at14d. The content of lignin and phenols kept increasing and the content of ligninincreased significantly at14d,22d and29d.4. During the healing process, soluble sugar content in healed graft union keptdecreasing but increasing in unhealed graft union. Soluble sugar content in unheald graftunion was significantly higher than the healed plants at22d. Phenols content in unheald graft union was significantly higher than that in heald graft union. The activities of POD, CAT andPPO, the content of H2O2, lignin, IAA in the graft uion were higher than that in rootstock.5. Increased acitivities of POD and CAT, IAA content and the accumulation of ligninare favorable to graft union development. The accumulation of phenols might be unfavorableto graft union development. The crucial time for healing process is two weeks after grafting.
