In this experiment two year old seedlings and five year old 'yanlong' Chinesechestnut trees were used as rootstocks and 'yanlong' branches as scions. The effects oflow temperature,water loss,different times and different methods on the rate ofgrafting survival were studied. At the same time, The influence of soluble sugar,starch,soluble protein,POD,SOD,CAT and Phenol compounds on the rate ofgrafting survival were analyzed. The results are as follows:1. The electrolyte leakage rate and the damage degree of chestnut branches wereincreasing with the temperature decreasing. There was a inflection point of thecontent of MDA and the activity of PPO at-20โ„ƒ, The rate of survival was0%under-20โ„ƒ.weinferred:(1)It was the lethal temperature at-20โ„ƒof 'yanlong' Chinesechestnut.(2)It was confirmed preliminarily that used the temperature appearing theinflexion to ensure the lethal temperature of plant.2. The rate of grafting survival was14.4%when materials were loosed water innatural conditions2d (rate of water loss7.12%, moisture content36.8%). The rate ofgrafting survival was0%when materials were loosed water in natural conditions4d(rate of water loss13.31%, moisture content32%).3. The rate of grafting survival trends were similar to two year old seedlings andfive year old Chinese chestnut trees. It was highest in April25th (The rate of graftingsurvival60%and63%). There had different survival rate in May25th,June25th,July25th and September25th. Scions can be gathered at different times on the basis,thereby the cost was reduced.4. The rate of grafting survival was positively correlated to its POD,SODactivity, the correlation coefficients between the rate of grafting survival and POD,SOD activity were0.9324,0.9892respectively, While the rate of grafting survivalwas negatively correlated to CAT activity, the correlation coefficients was-0.9676. The rate of grafting survival at different times were searched that could providetheoretical support for looking for grafting methods.5. There were three phenolic acid in two year old seedlings and five year oldrootstocks and scions. They were Gallic acid,4-hydroxy-benzoic acid and quercetin.There had minimal correlations between Gallic acid and4-hydroxy-benzoic acid andthe survival rate in two year old seedlings and five year old rootstocks and scions.While the rate of grafting survival was positively correlated to quercetin, thecorrelation coefficients were0.8802,0.9378,0.8746.
