Black poplar is a multipurpose and fast-growing tree species while its wood can be usedas pulp or artificial board material, also Commonly used for green planting. The selectionclone offspring is indispensable breeding engineering materials to poplar. Nearly30years,poplar breeding workers has slected a lot of black poplar clones for commercial cultivationand breeding research. However little differences could be found between large numbers ofthe clones, it is hard to Identificatate the clones by morphological characteristics. Clonesincorrectly named, This will seriously affect the Populus deltoides genetic research、varietiespromotion and protection and breeding efficiency. Traditional morphological identification isdifficult, time consuming and unaccurate.Isozyme and biochemical markers also show someobvious limitations. In this paper, we use SSR and ISSR molecular markers technology topreliminary build the six(including Lombardy poplar and the69)black poplar clonesfingerprinting, In order to provide theoretical basis for black poplar breeding research andvariety identification The results are as follows:1.Simple sequence repeat (SSR) was used to analyse the6Populus deltoides Genomepolymorphism, A total of10SSR primers which were polymorphic and informative wereselected to determine the genetic diversity and genetic relationships.132DNA bands wereamplified,125of which were polymorphic (94.67%).The size of Amplification bandsfragment ranged from100-1000bp.2.This research shows that using ISSR fingerprinting technique can accurately identifysix poplar clones,10ISSR primers were used for analying the poplar clones. We amplificateda total of82loci with53polymorphism loci.The percentage of similarity coefficient rangedfrom10.98%to69.51%. The average is8.2.3.ISSR and SSR Cluster analysis results all showed that Shansan and its female parent69gathered were clustered into the first group.Three Populus deltoides clones gathered forthe second category because of high genetic similarity.The third group contains onlyEuropean black poplar.The experimental results confirm the expectation. 4.ISSR bands from03-north1-15and Lombardy poplar were converted into twoSCAR markers which were used to rapid black Poplar clones identification.The results provereliability of the experiment.By through a large number of screening ISSR primers we canfound each black poplar clones the specificity SCAR markers, Further we can provide thetheory basis for rapid identification of black poplar material.
