The branch of two grape varieties “shuanghong”and“shuangyou” which belong to coldresistance Vitis amurensis were chosen as material, The influence of adventitious budsinduction, proliferation and rooting culture of “shuanghong”and“shuangyou” and thedetermination of the most appropriate matrix in transplanting stage were explored throughseveral aspects including the choice of explants, sterilization time and the ratio of the differenthormone; Then, the application of grafting in plant Vitis “shuanghong” tissue cultureseedlings leaves grapes for stock,with other frost-resistant different varietals tube seedlingsfor different varieties of the scion test-tube grafting,which aims at revealing frost-resistantdifferent varietals tube seedlings physiological and biochemical characteristics of thedifferences,and screening get frost-resistant better grape varieties and micro graftingcombination,for grape hardiness research and provide theoretical basis for cultivation.Research shows that:The tissue culture of vitis amurensis1.The sterilization effect was best when the stem of two Vitis amurensis varieties weredisinfected with75%alcohol first and then sterilized for8min in10%NaClO solution. Best“shuanghong”and“shuangyou”should be used explant length of about2.5~3.5cm,4~6mmin diameter around the upper single bud stems.2.The optimizing medium for proliferation of test tube plantlets was GS+NAA0.1mg/L,Value multiples can be achieved more higher.The best rooting medium was GS+IAA0.05mg/L, Allows up per plant the number of adventitious roots to9, The average length ofadventitious roots above5cm.3.The best matrix for transplanting plantlets was sand: perlite: vermiculite=1:1:2,Thestem of two varieties“shuanghong”and“shuangyou”Survival rate up to76.00%and70.00%.The micro-grafting of grapevines1.In the same total number, cleft grafting method grafted have the highest survival rate,Up to80%; other conditions are the same, The average survival rate that Single bud shoot tipand central stem, the lower single bud stems as scion was52.5%and39.98%,19.99%,so segments with tissue culture in vitro shoot tip grafting survival rate highest affinity best; Theaverage survival time of Vitis rootstocks “shuanghong” go to the root of21.88%, while leavetime rootstock grafted root the survival rate of39.38%, Choose to stay in vitro root graftingrootstock is more appropriate.2.Compared the survival rates among the four combination (red grapes/Vitis“shuanghong”,Phoenix/Vitis“shuanghong”,Pinot Noir/Vitis“shuanghong”,Cabernet Franc/Vitis“shuanghong”),the survivals of the combination(red grapes/Vitis “shuanghong”)werebetter than Phoenix/Vitis “shuanghong” and Pinot Noir/Vitis “shuanghong”,the poorest oneis the combination of Cabernet Franc/Vitis “shuanghong”.3.In vitro grafting grafted15days preceding the rapid growth of leaves and branches of alarge number of increase from15to30days within a period of growing well, but not as goodas the early, more than30days after the growth rate slows and stabilized. In the15days to30days the growth rate of the “shuanghong” rootstock roots than the growth rate from30days to45days of fast, root number increased significantly, from30days after grafting rootingslowdowns,45days and30days root of the number are Little difference.
