Objective and significance:In order to exploring a new small bowel anastomo-sis,we applied fibrin glue in the end-to-end intestinal anastomosis of dogs and con-trasted with conventional intestinal anastomosis.This study was to assessclinical ef-fects of fibrin glue used in intestinal anastomosis of dogs and investigate possiblemechanism of action in promoting healing of anastomotic,thus provide a theoreticalbasis for the veterinary clinical application of fibrin glue.Methods:30local health mongrel dogs were divided randomly into two groupswhich were used to do the experimental research on different end to end anastomo-sis:The experimental group:simple suture(single equidistant turn interrupted suture,6-pin)+fibrin glue. Control group:traditional suture (single turn interrupted suture,16-pin).Record bowel anastomosis time;in postoperative3d、5d、7d and14d and28d,3dog for each group were randomly selected to observe intestinal anastomoticmorphological and histology changes and evaluation degree of the anastomotic adhe-sions,measure the content of hydroxyproline、Caspase3、alkaline fibroblast growthfactor and vascuLar endothelial growth factor;the blasting pressure was measured inpostoperative14d and28d,and anastomotic diameter ratio was in28d. statisticsanalyzed.Results:The experimental group intestinal anastomosis average time (17.96min) was significantly less than the control group (24.29min).on28d postopera-tive, the anastomotic internal diameter ratio of experimental group was95%, com-pared with82%in the control group, the difference was significant (P <0.01),while14d and28d postoperative,the experimental group ABP (12.72kPa and17.08kPa)were significantly higher than that of the control group (9.98kPa and15.50kPa).Morphological and histology changes:Dogs have different degrees of post-operative adhesions,but the experimental group was significantly lighter than thecontrol group.Experimental groups:On3d postoperative,a large number of in- flammatory cells and a small amount of fibroblasts and capillaries in anastomotic,granulation tissue began to form.On7d postoperative,mucosa has the trend ofcrawling to contralateral,mucous layer began to heal together.Anastomosis wasfilled with granulation tissue,proliferated fibroblasts and inflammatory cells de-creased.On14d postoperative,mucosal regeneration basically covered the anasto-mosis;the collagen fiber connection appeared on both sides of anastomotic;granulation tissue capillaries and fibroblasts continued proliferation;collagen fibersincreased and arranged clutter;anastomotic was bright red.On28d postoperative,mucous layer covered completely;inflammatory cells disappeared;blood vesselsreduced significantly;collagen fibers arranged in neat rows;only a white line wasglimpsed in anastomotic.It s the first phase of healing.Control group:On3dpostoperative,a large number of inflammatory cells and red blood cells was showedin anastomotic gap.On7d postoperative,Poor mucosal layer of aggregate;inflammatory cells in anastomosis reduced;fibroblasts and capillaries began to ap-pear.On14d postoperative,granulation tissue proliferated extensively;portiongranulation tissue broke into the lumen;the collagen fibers started generating. On28d postoperative,granulation tissue filled the wound;mucous layer recovery wasincomplete;a large anastomosis visible scar;collagen fibers proliferated and basicneat,the anastomotic dents is obvious.Elisa test results show that postoperative change of caspase3in the experimentaland control groups both are increased first then decreased,bottomed out on5d and7d postoperative,and on7d and14d postoperative rised to the top (respectively of107.1ng/g and97.25ng/g);On3d,5d,7d and14d,postoperative,basic fi-broblast growth factor levels of the experimental group were higher, but on28d post-operative it s lower than the control group (0.31ng/g and0.38ng/g);On3d,5d,7d postoperative,vascular endothelial growth factor levels in the control groupshowed a downward trend, while the experimental group showed a rising trend, On14d、28d postoperative,the changes of content are stabilized,but it was higher in the experimental group thanthat of in control group.The hydroxyproline content,showed an upward trend both in experimental group and the control group,the con-tent of the experimental group at each time point were higher,but the content of3dand28d had no significant differences in postoperative (P>0.05), but on5d,14dand7d difference was significant (P <0.05).Conclusion:Applicating simple suture+fibrin glue anastomosis intestine,whichis Easy to operate and can shorten the anastomosis time. In addition it can reducecomplications and promote secretion of Caspase3、bFGF、VEGF,which acceleratehealing of intestinal anastomosis....
