The Role Of AMPK On The Development Of Stressed Oocyte

There are two negative reasons that stress could effect the process of breeding for mammals. :(1) the interference of ovulation;(2) effecting on the maturation of implanted oocyte. The process is influenced by the interaction of HPA axis and HPG axis.Breeding is affected by energy regulation, but the relationship between a variety of physiological approach has not been fully elucidated. Gonadotropin-releasing hormone(GnRH) neurons will regulate the release of GnRH by integrating many physiological signals, and may affect the regulation of energy about reproduction. GnRH neurons receive the signal of glucose concentration, regulate the signal pathways for energy through AMPK pathway. This study was to confirm the AMPK participates in the process of oocyte maturation, and the interaction between the AMPK and the glucocorticoids. Finally clarify the AMPK pathway will participate in the oocyte development.Set up the in-vitro culture system of swine oocyte, preparation the cell culture medium of the sow oocyte that used from M1 to maturation.In 39℃,5% CO2 concentration and saturated humidity under the conditions of cultivation. Using AMPK activator AICAR and inhibitor Compound C, processing oocytes, statistical the oocyte maturation rate;Using the cell culture medium with glucocorticoid developed oocytes, statistical the oocyte maturation rate;Adding glucocorticoid to cell culture medium, to activate or inhibit AMPK signals respectively, detecting the change of oocyte maturation rate.Heat stress treatment was given to mice of eight weeks age, and the related effects of the same period of estrus and superovulation treatment of mice with heat stress on reproductive system, AMPK pathway in hypothalamus,the influence of blood indicators,the rapeutic effect on those of using drugs(bile acid).Two surveys about the sow fertility were taken in Shandong province, one is mainly aimed at various scale of sow farm,in order to investigate current status of the sow breeding in Shandong province and sow fertility deeply touched,by making questionnaire;the other is research on breeding group farms in the province by field trips.The purpose is to find out the effects of environmental factors on sow fertility and solutions through the investigation.
