Inner Mongolian Cashmere Goats was the valuable species of the world and the cashmere was the wellknow "cashmere diamond".The study showed that melatonin could promote the cashmere production,while the mechanism of the regulation and the signalling was still unclear.So,to explore the mRNA expression of melatonin acceptors, MTNRla.MTNRlb and RORαin follicles of February,April,August,October and December and find the regulation mechanism of the melatonin and signal pathway of melatonin by RT-PCR,fluorescene real-time quantitive PCR and in situ hybridization.The results showed:First,the MTNRla exon was composed of 1081 nucleotides and 345 amino acids were possibly encoded.A relatively high homology of the nucleotide sequences was found with other mammals,but it was low among different species.Secondly,RT-PCR indicated that there was no expression of MTNRla mRNA in cashmere goat skin tissues,but there was in the hypothalamus.There was no expression of MTNRlb mRNA in both skin and hypothalamus and RORαmRNA was expressed in the skin.Thirdly,in situ hybridization showed there was no MTNRla mRNA expression in the skin but expressed in hypothalamus.RORαmRNA was expressed in the skin and the expression was relatively high in cashmere shaft in February.The expression was mainly in the external root sheath,medulla and the cashmere shaft in April,and it was stronger in stem than follicle.There was relatively high expression in cortex and external sheath in August.It was quite the same in October as it was in February in the hair shaft.And the expression was found in the medulla and the internal root sheath in December.Fourthly,fluorescene RT-PCR showed that RORαmRNA was expressed in the skin in February,April,August,October and December,the highest expressed was in February, lower in April,rised in August and October and achieved the level in October as it did in February,and got to the lowest in December.It showed kinetic change.Finally,the expression of the RORαmRNA was mainly in the hair shaft in the skin heelinned with the melatonin,and the expression was relatively low compared with the common tissue.The expression of RORαmRNA was detected in the skin follicle of Inner Mongolian Cashmere Goats and the expression of different months were kinetic change in this study. It showed the way of melatonin promoting the cashmere production probably because of melatonin acceptors.It provided the basis to fouther study the relationshipof melatonin acceptors and cashmere production,explain the regulation mechanism and signal pathway of melatonin and manual control cashmere production.
