Dastarcus helophoroides Fairmaire(Coleoptera: Bothrideridae) is the most effective natural enemy against many longhorned beetles. In addition, the adult Dastarcus helophoroi des which has a relative long lifespan could live several years long under laboratory condition. The oxidation-reduction reaction in the aerobic organisms can produce a large amount of Reactive oxygen species(ROS) like O2-. If the excessive ROS could not be removed in time, it will cause damage to the macromolecules like proteins in the cell, which could further cause diseases. The organisms have developed a complete defence system to remove it during the accumulation of ROS, including the antioxidant enzyme system such as SOD, CAT and Prx. In order to explore the possible roles that the antioxidant enzymes play in the long lifespan of Dastarcus helophoroides, we measure the expression level of SOD and Prx6 transcripts using RT-qPCR. The main results are as follow:There were 5 SOD, 3 CAT and 5 Prxs transcripts in the Dastarcus helophoroides transcriptome dataset. After online Blastx analysis, the 5 SOD transcripts were classied into 3 different subgroups: Cytoplasm Cu/Zn-SOD, mitochondrial Mn-SOD and Extracellular Cu/Zn-SOD. The 5 Prxs transcripts were classified into two groups: 3 Prxs were in 2-Cys subgroup and 2 in 1-Cys subgroup.The full-length of D.hSOD2 and D.hCAT1 were cloned with RACE-PCR. Their open reading frames(ORF) were 642 bp and 1536 bp, coding 214 and 512 aa respectively. All the active sites, conserved motifs and signal aequences of SOD, CAT and Prxs were analysed. The phylogenic analysis showed they share the high homology with that of Tribolium castaneum.The RT-qPCR results showed all the SOD and Prx6 transcripts can be detected in all the examined tissues, but exhibited tissue-specific. In addition, their expression fluctuates during development but showed relative expression level during different age groups of adults after eclosion.
